醋酸棉酚对小鼠黄体细胞凋亡的影响

【摘要】 【目的】为了探讨醋酸棉酚对小鼠黄体细胞凋亡的影响;【方法】将25-30日龄昆明系雌性小鼠80只,称重后随机分为5组,分别为空白对照组,乙醇对照组,染毒低、中、高剂量组,每组16只。取卵巢黄体进行以下研究:黄体组织切片观察,卵巢黄体MDA含量和SOD活性及血液中孕激素的含量的测定,Tunel法检测小鼠卵巢黄体细胞凋亡,RT-PCR法测定Bcl-2,Bax和P53基因表达:【结果】醋酸棉酚可以直接损伤小鼠黄体细胞,产生明显的细胞毒性、且这种毒性作用呈正相关;SOD活性与孕激素的含量随着棉酚剂量的增高而降低,而MDA含量变化趋势与此相反。各染毒组小鼠的卵巢黄体细胞凋亡率明显升高,与对照组相比差异极显著(P<0.01)。Bax和P53基因表达水平明显上升,Bcl-2基因表达水平明显下降;【结论】棉酚能有效地诱导小鼠黄体细胞发生凋亡。
【Abstract】 To approach the Effects of gossypol induced luteal cells apoptotic in Mice.;The kuming mice will be 25-30 days old female mice of 80,after weighing were divided into five groups,the control group,ethanol control group,gossypol to low,middle and high dose groups, corepus luteum was taken for following research:corepus luteum tissue observation,MDA content and SOD activity and progesterone Determination in blood,detection corpus luteum cell apoptosis by Tunel,BCL-2,Bax gene and P53 expression by RT-PCR;Gossypol can kill mice luteal cells,resulting in significant cytotoxicity and a positive correlation.With the degrading the dose of gossypol,SOD activity and progesterone levels increased,while the MDA content trends to the contrary,luteum cell apoptosis was significantly higher compared with the control group significant deviation(P <0.01).Bax gene and P53 gene expression levels were increased obviously,Bcl-2 gene expression was reduced.
