不同钼水平对山羊红细胞膜自由基代谢及其酶活性的影响

【摘要】 (目的)本文旨在观察不同钼水平对山羊红细胞膜自由基代谢及其酶活性的影响。(方法):选用27头健康山羊随机分成3组,选用钼酸铵[(NH4)6Mo7O244H2O]作为试验钼源,Ⅰ组为对照组,Ⅱ组钼15mg·kg-1,Ⅲ组钼45mg·kg-1。试验50天,并于第0、10、20、30、40、50天采血,测定红细胞膜中自由基含量和酶活性变化。(结果):组内比较,Ⅱ组、Ⅲ组GSH-Px、T-AOC、CAT、GST、T-AOC、ATPase和AKP活性在试验前期(0~30天)差异不显著(P>0.05),实验后期(40~50天)与前期比较下降,差异显著(P<0.05);MDA含量、XOD和LDH活性在试验前期(0~30天)差异不显著(P>0.05),后期(40~50天)与前期比较升高,差异显著(P<0.05)。组内比较,Ⅱ组、Ⅲ组GSH-Px、T-AOC、CAT、GST、T-AOC、ATPase和AKP活性在试验前期(0~30天)与对照组比较差异不显著(P>0.05),后期(40~50天)与对照组比较降低,差异显著(P<0.05);MDA含量、XOD和LDH活性前期(0~30天)与对照组比较差异不显著(P>0.05),后期(40~50天)升高,差异显著(P<0.05)。(结论):由此可见,山羊长期摄入过量的钼会导致红细胞膜抗氧化功能降低、自由基含量升高和能量代谢障碍,受损红细胞增加,红细胞寿命减短。
【Abstract】 (Objective) This experiment was conducted to determine the effect of different levels of molybdenum on the metabolism of free radical and activities of enzyme in erythrocyte membrane from goats.(Method) 27 goats were randomly assigned to three different treatments.Treatments consisted of control(no additional Mo,groupⅠ) and high molybdenum (Mo 15mg·kg-1,groupⅡ:Mo 45mg·kg-1,groupⅢ) from[(NH4)6Mo7O24·4H2O].The goats were reared until 50 days old under normal condition.Blood samples were collected on days 0,10,20,30,40 and 50d to determine the metabolism of free radical and activities of enzyme in erythrocyte membrane from goats.(Result) The activities of the enzymes GSH-Px, T-SOD,CAT,GST,T-AOC,ATPase and AKP were significantly decreased(P<0.05) of high molybdenum groups after 40 day:the MDA contents,XOD and LDH activities were significantly increased(P<0.05) of high molybdenum groups after 40 day.When the control group was compared with each of the two high molybdenum group,the activities of the GSH-Px, T-SOD,CAT,GST,T-AOC,ATPase and AKP were significantly decreased(P<0.05) appeaered for groupⅡand groupⅢduring days 40~50.the MDA contents,XOD and LDH activities were significantly increased(P<0.05) in high Mo groupsⅡandⅢthan that in control group from 40 day onwards.(Conclusion) The results showed that high molybdenum impaired antioxidant’ s function,increased free radical contents and obstructed energy metabolization of enzyme in erythrocyte membrane from goats.The function of erythrocyte membrane was finally injuried.The injuried erythrocyte was increased.The lifetime of erythrocyte was decreased.
