汉族健康大学生群体中COMT Val158Met功能多态性对工作记忆和双侧海马体积的作用

Impact of Functional Variant in COMT(rs4680) on Working Memory and Bilateral Hippocampus Volumes in Healthy Chinese College Students

【摘要】 COMT Val158Met(rs4680)基因多态性对调节大脑多巴胺水平具有关键作用,而后者又进而调节脑发育过程和工作记忆的容量。已有研究证实rs4680多态性与个体工作记忆容量相关并且具有种族差异,同时rs4680多态性对海马等与工作记忆相关的脑区具有作用,但研究结果局限于高加索裔群体,缺乏亚裔被试研究。因此,本研究考察了rs4680多态性与19到21岁中国汉族健康大学生的工作记忆(N=446)以及海马体积(N=331)之间的联系。以2-back范式为任务对工作记忆进行测查,并利用西门子3-T磁共振扫描仪收集脑结构数据(T1像),用freeSurfer软件分割和测量双侧海马的体积,并通过PCR测查rs4680的基因型。线性回归分析发现,随着rs4680Met等位基因的数目增加,工作记忆正确率(性别为协变量)和双侧海马体积(性别和颅内体积为协变量)都出现显著的下降趋势;而双侧海马体积随着工作记忆正确率上升而显著增加。该结果与高加索群体的结果呈相反趋势。该研究为COMT Val158Met对工作记忆和海马体积的作用增加了佐证。

【Abstract】 COMT Vall58Met(rs4680) polymorphism plays a crucial role in regulating the dopamine level in the brain,which in turn modulates the development of brain and working memory(WM) capacity.It is reported that the function of rs4680 on WM seems to vary among different races at the behavioral level.Previous studies showed that rs4680 affects brain regions related to WM(e.g., hippocampus);but most of them focused on Caucasian population while little is known about Asian samples.The current study investigated the association of COMT Vall58Met genotype with working memory(iV = 446) and hippocampus volume(N = 331).All participants were healthy Han Chinese college students(age 19 to 21).WM was assessed with typical 2-back paradigm.Tl-MR image data were collected from a Siemens 3-T Magnetic Resonance Scanner,volume of hippocampus was segmented and measured with freeSurfer package.COMT Vall58Met polymorphism was genotyped with standard PCR.Linear regression analyses showed the number of the COMT Met allele was associated with the significant decrease in both WM accuracy(gender as covariate) and bilateral hippocampus volume(gender and intracranial volume as covariate).The relation between WM accuracy and bilateral hippocampus volume was significandy positive when considering gender.This pattern is opposite to the previous findings for Caucasian samples.The results add to the growing literature about the effects of COMT rs4680 polymorphism on WM and hippocampus volume in Chinese Han populations.
