【摘要】 目的比较结核病患者与健康人血清蛋白质谱,寻找结核病相关特异血清蛋白质,以期发现新型结核病血清学诊断标志物。方法应用WCX蛋白质芯片和表面增强激光解吸-电离飞行时间质谱(SELDI-TOF-MS)技术对包括54例菌阳肺结核(菌阳组)、45例菌阴肺结核(菌阴组)、18例肺外结核(肺外组)共117例结核病患者(结核组)及62例健康对照(对照组)进行了血清蛋白质谱检测,并应用ClinProTools分析软件进行分析,找出差异蛋白质。结果菌阳组-对照组、菌阴组-对照组、肺外组-对照组和结核组-对照组间分别存在19、14、26和30个差异血清蛋白质,其中分别有11、9、15和15个蛋白质在病例血清中表达上调;质荷比(m/z)分别为1060、1944、2081和3954的4个血清蛋白质为各患者组与对照组间共同存在的差异血清蛋白质,其中m/z为1060的蛋白质在病例血清中表达下调。结论结核病患者与健康入之间血清蛋白质谱存在差异,有望通过血清蛋白质谱的比较分析寻找到结核病相关特异血清蛋白质而发现新型结核病血清学诊断标志物。但尚需大样本量、多对照系的进一步研究予以确证。更多还原

【Abstract】 Objective To look for specific tuberculosis-related serum proteins and further to discover new serological diagnostic markers of tuberculosis by comparison of the serum protein spectrum between tuberculosis patients and healthy people.Methods The serum protein spectra from 62 healthy controls (control group) and 117 cases of tuberculosis patients(TB group),including 54 cases of sputum or culture positive pulmonary tuberculosis(bacterial positive group) and 45 cases of sputum and culture negative pulmonary tuberculosis (bacterial negative group) and 18 cases of extra-pulmonary tuberculosis (extra-pulmonary group),were detected using the WCX protein-chip and the surface-enhanced laser desorption ionization time of flight mass spectrometry (SELDI-TOF-MS) technology.Then the detected data of serum protein spectra were analyzed by ClinProTools Software to find the difference proteins. Results 19,14,26 and 30 protein peaks were significantly different(P<0.05) respectively in bacterial positive group vs.control group,bacterial negative group vs.control group,extra-pulmonary group vs.control group,and TB group vs.control group.11,9,15 and 15 protein expression of them were up-regulated in kinds of tuberculosis 7patient groups.Four protein peaks at 1060,1944,2081 and 3954 m/z were co-existing different proteins between kinds of tuberculosis patient groups and control group,of which 1060 m/z protein was relative low expression in kinds of tuberculosis patient groups. Conclusions There are difference of the serum protein spectrum between tuberculosis patients and healthy people.New serological diagnostic markers of tuberculosis may be discover by researching specific tuberculosis-related serum proteins.But,a study of large scales samples and multi-control system need to perform in order to obtain reliable conclusion.更多还原
