Secretome of perivascular adipose tissue:The affects of complement C3 on adventitial fibroblast

【摘要】 <正>Background:Perivascular Adipose Tissue(PVAT)is an important component of blood vessels.The cytokines secreted from adipose tissue are considered to play critical role in regulation of vascular function.However,the effects of cytokines released from PVAT on vascular adventitial fibroblasts(AFs)have not yet been investigated.In this study,we investigated whether PVAT-conditioned medium(PVATCM)could affect cellular function of cultured rat aortic AFs.Methods:The PVATCM was collected from the aorta of SD rats,we identified the secretome of PVAT utilizing a liquid chromatography-tandem mass spectrometry(LC-MS/MS)method and classified these proteins into groups according to their functions in cardiovascular system.We also verified the expression and function of complement component C3 on the AFs.Results:Our results showed that PVAT-conditioned medium stimulated migration and inducedα-smooth muscle actin expression,which was used as a characterized marker of fibroblasts-to-myofibroblasts differentiation.Furthermore,Complement C3 was considered to be abundantly secreted from PVAT according to the proteomic strategy.We found that the affects of PVATCM were attenuated after blocking the C3,we also found that exogenous C3 stimulated the migration and differentiation of AFs.Conclusion:Our study demonstrated that PVAT-conditioned medium stimulated migration and differentiation of AFs.And we firstly identified the secretome of PVAT using a proteomic method,which supported the paracrine role of PVAT in pathophysiological states through the secretion of cytokines.We also demonstrated that C3 play an important role in AFs migration and differentiation.
