【摘要】 为了对家蚕Bombyx mori L.精子介导转基因技术进行探索,将转基因载体与表达转座酶的辅助质粒混合后,注射进处女蛾的交配囊内,交配后产下卵即时浸酸,正常催青,用荧光显微镜观察蚕卵的荧光情况,发现有部分蚕卵发出绿色荧光,正常饲养至化蛾,荧光显微镜观察进行筛选,发现有12头家蚕蛾表现出明显的荧光,荧光蚕蛾PCR检测扩增出EGFP基因的特异性条带。Southern杂交分析表明荧光蚕蛾的基因组中含有EGFP融合序列,表明所获得的荧光蚕为转基因家蚕。更多还原

【Abstract】 In order to explore the method of sperm-mediated transgenic Bombyx mori L.,transposon vectors DNA were injected into a virg in moth s bursa compulatrix.Then,the treated moths mated with normal males and laid eggs naturely.Green fluorescent were observed in 5%in of eggs.The eggs were prossed and reared with the normal way,and twelve moths were observed with green fluorescent.PCR and Southern blot assay showed that EGFP fragments were successful intergrated into the chromosome of silkworm.更多还原
