视黄酸受体及类视黄醇X受体激动剂对T细胞分化及受体基因表达的影响

【摘要】 目的:研究视黄酸受体和类视黄醇X受体激动剂在体外对T淋巴细胞凋亡、发育、表面CD标志的分化及核内视黄酸受体亚型表达的影响。方法:采用PI单染流式细胞术分别测定不同浓度视黄酸受体激动剂TTNPB和类视黄醇X受体激动剂MA处理T淋巴细胞后的凋亡情况。以FITC、PE双染流式细胞术检测T淋巴细胞CD4+和CD8+细胞的比例。以ELISA法检测培养上清液中IL-4和IFN-γ含量。用实时荧光定量PCR检测TTNPB和MA对脐血T淋巴细胞视黄酸和类视黄醇X受体亚型基因的转录调节作用。结果:100μM的TTNPB可以使T淋巴细胞凋亡率有所增加。10μM的MA在体外能降低CD8+T细胞比例,从而提升CD4+/CD8+比值,而RARs激动剂TTNPB各组和对照组之间差异无显著性。MA能在体外抑制T淋巴细胞IFN-γ分泌,同时促进T淋巴细胞IL-4的分泌。10μM和100μM的TTNPB能诱导T淋巴细胞RARαmRNA表达。MA能抑制T淋巴细胞RARαmRNA和RXRαmRNA的表达。结论:RXRs激动剂MA能调节T淋巴细胞发育和分化,RARs及RXRs激动剂均能调节视黄酸受体亚型的表达。RA调节T细胞免疫的功能的作用可能是由视黄酸及类视黄醛X受体途径共同作用下实现的。

【Abstract】 Object:To investigate the effect of RARs and RXRs agonists on the apoptosis,development,differentiation and the espression of RA receptors subtype. Methods:Purified T cells were cultured with indicated concentrations of RARs agonist TTNPB or RXRs agonist MA(0.1、1、 10、100μM) for 48h,respectively. Apoptosis and the propotions of CD4+ and CD8+ T cell were detected by FCM respectively. The levels of IL-4 and IFN-γin supernatant were quantitated by ELISA. The expression of RARs and RXRs subtypes were analyzed with RT-Q-PCR. Results:T cell apoptosis rate was promoted by TTNPB at 100μM. The propotion of CD8+ T cell was reduced by MA at 10μM,yet TTNPB has no such effect. In the MA groups IFN-γconcentration were significantly lower than the controls,while IL-4 concentration were significantly greater. Additionally,RARαmRNA was induced by TTNPB at 10μM and 100μM,and RARαmRNA 和 RXRαmRNA were suppressed by MA. Conlusion:The RXRs agonists MA can regulate T cell immune fuctions in vitro. besides,TTNPB and MA can adjust the expression of RA receptor subtypes. The effects of RA on T cell immune fuction was probably achieved by the combined action of RARs pathway and RXRs pathway.
