三丁基锡对杂色鲍若干基因mRNA表达的影响

【摘要】 杂色鲍(Haliotis diversicolor)是福建省海水养殖贝类重要品种之一,但在养殖业迅速发展的同时野生鲍资源已受到严重破坏,有研究表明这与三丁基锡(tributyltin,TBT)在其体内累积诱发性畸变而造成种群的下降密切相关。本研究采集1μg/g(wt)TBT暴露28d、56d后的样品,采用生物信息学软件分析并筛选获得鲍应激诱导相关基因,利用real-timeQPCR方法分析CaM、GST、CytIII等37个基因的差异表达特性,证实受TBT诱导肝胰腺及性腺内AP-4、Apoptosis等20个基因显著的上调表达,MIP-I-exon、Ubi、GST等3个基因显著的下调表达,HMG1、Cytidine等14个基因波动表达。这些结果有助于进一步了解TBT导致海洋腹足类性畸变的分子机制,获得的应激诱导相关基因还可作为特异的分子标记为TBT及其它内分泌干扰物的早期快速监测提供可能。
【Abstract】 Haliotis diversicolor is one of the commercially important shellfish species for aquaculture in Fujian province, but its wild resources have been severely damaged since rapid economic development. Some studies indicated that the population decline was closely associated with imposex induced by tributyltin (TBT) accumulation in body. In present study, 37 genes including CaM, GST, CytIII were chosen and the changes of mRNA levels after exposed to 1 μg/g(wt) TBT from hepatopancreas and gonad were studied using real-time Q PCR. The results show that, 20 genes including AP-4, Apoptosis were up-regulated, 3 genes (MIP-I-exon, Ubi, GST) were down-regulated, while 14 genes like HMG1, Cytidine were differently induced in different tissues or at different exposue time course. These results can help us further understand the molecular mechanism of marine gastropods imposex induced by TBT, and the obtained genes related to gonad development and stress response can also be used as specific biomarker candidates for TBT and other endocrine disrupting chemicals monitoring.
