【摘要】 为确定泰山赤鳞鱼(Varicorhinus sp.)的分类地位,本文用核内与核外DNA分析了泰山赤鳞鱼与多鳞铲颌鱼(Varicorhinus macrolepis)的关系。基于cytb和D-Loop序列发现泰山赤鳞鱼和多鳞铲颌鱼之间的遗传距离分别是0.1%～1.0%和0～0.5%,与泰山赤鳞鱼和多鳞铲颌鱼个体之间的遗传距离几乎一样。从cytb和D-Loop聚类图上都可明显看出二者聚在一起,没有任何分别。利用5对微卫星引物进行扩增,分析发现两个群体遗传距离非常小,为0.0113,而遗传相似性高达0.9887。因此,无论是cytb、D-loop全序列分析,还是微卫星标记分析均可看出泰山赤鳞鱼和多鳞铲颌鱼是同种,即泰山赤鳞鱼的学名就是多鳞铲颌鱼(Varicorhinus macrolepis)。更多还原

【Abstract】 To carify the taxonomic status of Varicorhinus sp., the relationship between Varicorhinus sp. and Varicorhinus macrolepis was analyzed by using nuclear and mt DNA. Based on the whole sequence of cytb and D-loop, the genetic distance between Varicorhinus sp. and Varicorhinus macrolepis were 0～0.5% and 0.1%～0.6% respectively, which were same almostly to the genetic distance between individual of Varicorhinus sp. and Varicorhinus macrolepis. Phylogenetic tree based on cytb and D-loop sequences also showed that the Varicorhinus sp. and Varicorhinus macrolepis were clustered together. The results demonstrated they belong to the same species. In addition,we also analyzed the gene frequency, genetic distance and genetic similarity of the two group using microsatellite marker. The result revealed that the polymorphism of Varicorhinus sp. and Varicorhinus macrolepis was very poor,the genetic distance between two group was vey small (0.0113), and the genetic similarity was high (1.9887). Therefore, based on mtDNA cytb, D-loop and microsatellite sequences, we conclude that Varicorhinus sp. and Varicorhinus macrolepis belong to one species.更多还原
