Cu2+胁迫对禾花鲤(Procypris mterus)过氧化氢酶活性的影响

【摘要】 本研究旨在探讨Cu2+浓度梯度、胁迫时间对禾花鲤(Procypris mterus)血液和肝脏的过氧化氢酶(Catalase,CAT)活性的影响,为禾花鲤的重金属胁迫研究及水污染生物监测提供理论参考依据,进一步指导禾花鲤的健康养殖。采用可见光法测定了不同Cu2+浓度(0mg/L、0.067mg/L、0.084mg/L、0.101mg/L、0.118mg/L、0.134mg/L)和胁迫时间(0d、1d、5d、11d)下禾花鲤CAT活性的变化。结果显示,在试验的各个持续时间段(1d、5d、11d),低Cu2+浓度处理组(0.067mg/L)血液及肝脏的CAT活性高于其它各处理组,且活性随着胁迫时间的延长而增加。而中、高Cu2+浓度处理组(0.084mg/L、0.10lmg/L、0.118mg/L、0.134mg/L)血液和肝脏中的CAT活性则随着Cu2+浓度的升高和胁迫时间的延长被显著抑制(P<0.05),活性变化在5d后趋于稳定。试验表明鱼体过氧化氢酶的活性受水体中Cu2+浓度的影响,低Cu2+浓度能诱导鱼体血液和肝脏中的CAT活性增加,但随胁迫的加剧,CAT活性受到抑制。

【Abstract】 The aim of this experiment was to examine the effects of copper stress on the catalase(CAT) activity in blood and liver tissue of Carp(Procypris mterus).Carps were treated with 0mg/L、0.067mg/L、0.084mg /L、0.101mg/ L、0.118mg/L、0.134mg/L of Cu2+ for periods of 0 to 11d under visible light condition.The results showed that highest CAT activities in blood and liver tissue were observed in the lower Cu2+ concentrations treatment(0.067mg/L) and CAT activities of this treatment were increased with exposure time.However,CAT activities in blood and liver tissue in other treatments(0.084mg/L、0.101 mg/ L、0.118mg/L、0.134mg/L Cu2+ concentrations) were decreased with Cu2+ concentrations and exposure time (P<0.05) and tended to be stable after 5 days.The experiment suggests that CAT activity can be affected by Cu2+ concentrations in water,and a low dose stimulation followed by a high dose inhibition.
