Pim-3反义寡核苷酸对人肝癌细胞SMMC-7721生物学影响

【摘要】 目的研究Pim-3硫代反义寡核苷酸(Pim-3 ASODN)对SMMC-7721肝癌细胞生物学影响及其作用机制。方法通过脂质体介导Pim-3 ASODN转染SMMC-7721细胞,噻唑蓝(MTT)法检测细胞增殖,逆转录-聚合酶链反应(RT-PCR)方法检测Pim-3 mRNA的表达,Western blot检测细胞Pim-3蛋白的表达,结果 MTT法检测显示:与错义ODN、脂质体和DMEM培养基的三组肝癌细胞相比,Pim-3 ASODN明显抑制SMMC-7721细胞增殖,且呈时间和剂量依赖性,差异有统计学意义(P<0.05);RT-PCR、Western blot检测显示与3对照组相比,转染了Pim-3 ASODN的细胞mRNA和蛋白表达明显减少,差异有统计学意义(P<0.05)。结论 Pim-3 ASODN能够明显抑制SMMC-7721细胞增殖。Pim-3 ASODN有可能成为肝癌基因治疗的一个新靶点。
【Abstract】 Objective:To investigate biological effects of Pim-3 fully phosporothioated antisense oligodeoxynucleotide(Pim-3) ASODN uman hepatocellular carcinoma SMMC-7721 cell.Methods:Pim-3 ASODN was transfected into SMMC-7721 cells by Lipofectamine 2000.Cell proliferation was detected by MTT.The expression of Pim-3 was detected by Reverse Transcription—Polymerase Chain Reaction (RT-PCR) and Western blot.Results:MTT assay demonstrated that Pim-3 ASODN could significantly inhibit the proliferation of the cells in both time and concentration dependent manner,compared to missense ODN,liposomes and DMEM medium(P<0.05).RT-PCR and Western blot analysis showed the mRNA and protein expression of Pim-3 ASODN cells compared with three control groups was significantly reduced (F<0.05).Conclusion:Pim-3 ASODN is able to inhibit the proliferation of SMMC-7721 cell,which might provide a new path for hepatocellular carcinoma gene therapy.
