【摘要】 目的乙脑疫苗是我国基础免疫的主要生物制品之一,乙脑病毒感染人体后遗症严重,死亡率也较高,而注射疫苗是唯一有效的预防方法。乙脑疫苗的生产工艺经历了鸡胚培养疫苗、鼠脑疫苗、原代地鼠肾灭活疫苗和原代地鼠肾减毒活疫苗,以及少量的Vero细胞纯化灭活疫苗。目前,在国内多使用原代地鼠肾减毒活疫苗,该疫苗的注射剂最小副反应少,也具有较好的安全性和有效性,但在制备过程中,由于使用的地鼠肾原代细胞,这就存在一个高污染的问题以及受到动物源性的各种不明外源因子的影响。为了减少污染及外源因子的影响,制备出具有更好免疫性、更安全和更经济的乙脑疫苗,我们尝试着使用Vero细胞来制备纯化乙脑减毒活疫苗。经检测,纯化后的疫苗的免疫性和安全性良好。因此,用这种方法和材料有望生产出一种更安全和有效的乙脑减毒活疫苗。更多还原

【Abstract】 In order to produce a second generation Japanese encephalitis(JE) Vaccine in Vero cell that is safe,effective,and possibly more economical.The JE PAV was made from an attenuated strain of JE virus propagated in certified Vero cell,.Its manufacture followed current GMP guidelines for the Production of biologicals.The manufacturing process was efficient in generating a high yield of virus essentially free of contaminating host cell protein and nucleic acids.Vaccinated animals developed high-titered JE virus neutralizing antibodies in a dose dependant fashion after one or two injections.The vaccine protected mice against morbidity and mortality after challenge with live.virulent,JE virus.Compared with the existing vaccine JE-Vax,the cell-derived JE-PAV was more immunogenic and as effective.as preventing encephalitis.更多还原
