东方百合根尖染色体制片技术的研究

【摘要】 以东方百合‘Montezuma’为材料,研究了根尖的取材位置、不同预处理液和解离液处理对染色体制片技术的影响,并对染色体的数目进行了观察。结果表明:在茎生根长出后,茎生根分裂旺盛程度比基生根好,分裂相较多,适宜用做染色体制片材料。冰水混合物48h进行预处理时,染色体收缩程度高,分散好,缢痕明显,中期分裂细胞较多,可以用于染色核型以及分带研究;0.05%的秋水仙素预处理3h效果次之,染色体形态清晰,分散程度较高,收缩程度高,中期分裂细胞较多,适宜用于染色体计数研究。15%HCl:95%乙醇=1:1制成的混合液室温(25℃)解离5min时,根尖就能够得到充分的软化,压片时根尖易于分散,镜检时细胞分散程度好,且不易压碎,效果最好,解离8min的效果与之相差不大。最后对材料进行染色体计数,此品种为二倍体,染色体数目2n=2x=24。

【Abstract】 The roots of Oriental lily‘Montezuma’ were used to study the effects of the sampling position, the pretreatment chemical solution and the dissociation solution,the number of chromosomes were observed. The results showed that the mitotic division of the stem-roots were more vigorous after they appeared,the stem-roots were more suitable to be the chromosome squashing material.Among the four pretreatment methods, the material in ice water mixture for 48 hours was optimal to karyotype analysis and chromosome banding research,the chromosomes got were well contracted,uniformly dispersed and the cells in metosis metaphase were more.The material in 0.05%colchicine solution for 3 hours had a better effect in the remainings,the chromosome morphology was clear,the contraction was short,the method was more suitable for chromosome counting research.The mixture of 15%HCl and 95%ethanol can dissociate the root tip well,it made the root tip cells disperse easily,during the microscopic examination,the chromosome dispersed evenly,the effect was better than the other dissociation solutions.After counted the chromosome number of 30 cells,the results showed that the ’ Montezuma’ is diploid(2n=2x=24).
