Cry1Ac基因在玉米中的转化与表达

【摘要】 本文对采用基因枪介导法获得的转BT基因玉米植株进行PCR鉴定和Southern检测,在DNA水平上证实目的基因Cry1Ac-w成功转入玉米植株体内,同时又对其进行了BT蛋白试纸条检测和ELISA分析,在蛋白水平上检测分析了其在Z58Ac-w1和Z58Ac-w3两种转化体不同生育时期各组织器官的蛋白表达量。结果表明:同一生育育期内不同组织器官cry1Ac的蛋白表达量不同,差异显著,叶片蛋白表达量最高达153.3ng/g,根最低;不同生育期,cry1Ac在叶片中的表达量不同,差异显著,各生育期BT蛋白表达量顺序:抽穗期>灌浆期>成熟期。

【Abstract】 Identification of transgenic plants by PCR detection and southern blot analysis revealed that the Cry 1 Ac - w had integrated in the plant genome.Simultaneously,protein expression of transgenic plants of different transformation,different growth stages and different organs were tested respectively by BT protein Lateral Flow strip and ELISA.Results showed that the protein expression vary among different transformed in the same growth stage,Z58Ac - w1 is higher than Z58Ac - w3 in the average expression,the difference is not significant;protein expression of different tissues and organs in the same reproductive fertility are different,blade and leaf sheath is as high as 153.3ng/g,while root at the minimum;Cry1Ac expression in leaves of different growth stages are different on the decline,furthermore the difference is significance.The order of protein expression in different growth stages;heading stage>filling>maturity.
