转Cry9HNt基因玉米植株的获得及抗虫性鉴定

【摘要】 以玉米杂交组合PA×PB的胚性愈伤组织为材料,通过农杆菌介导法将本实验室人工改造的Cry9HNt抗虫基因导入到了玉米基因组中,并对获得的转基因植株进行PCR鉴定,进而选取部分PCR呈阳性的植株进行Southern杂交,结果表明Cry9HNt基因已经整合到玉米基因组中。BT蛋白免疫试纸鉴定结果也进一步证实了目的蛋白的表达。对转基因T1代植株进行田间抗虫鉴定,结果表明转基因玉米植株的抗虫活性显著地优于非转基因对照,这也进一步证实了我们改造完成的Cry9HNt基因具有良好的抗虫活性。
【Abstract】 Artificially transformed Cry9HNt insect-resistant genes were transferred into embryogenic maize callus of hybrid line PA X PB by agrobacterium tumefactions-mediated transformation approach.After selection,transgenic plants were regenerated,and some of them were positive by PCR and part of the PCR positive selection of southern plants hybrid,the results show that Cry9HNt gene has been integrated into the maize in the genome.BT protein immune test appraisal result also further confirmed that the purpose of protein expression.in the transgenic plants T1 generation plants field insect resistance identification,and the results show that genetically modified corn plant resistance to insects activity significantly better than non-modified comparison,this also further proves that we transform Cry9HNt has good resistance to insects activity.
