针刀松解法与电针对膝骨关节炎大鼠中枢β-EP及CCK-8的影响
Effect of Acupotomy Lysis and Electro-Acupuncture on β-EP and CCK-8 in Central Nervous System of Rats with Knee Osteoarthritis

【摘要】 目的:对比观察针刀松解法及电针对膝骨关节炎大鼠模型中枢疼痛相关神经肽的影响。方法:将80只健康SD大鼠随机分为空白组、模型组、针刀组和电针组。将4%木瓜蛋白酶溶液与0.3mol/L半胱氨酸溶液按1:1混合静置0.5h后分别于造模第1、4、7d注射于模型组、针刀组和电针组大鼠左膝关节腔,每次20μL。4周后相应给予针刀松解治疗或电针治疗。治疗3周后取大鼠腰膨大处脊髓及中脑,分别测定β-EP及CCK-8的含量。结果:模鼠患膝关节软骨暗淡发黄、部分软骨象牙化、有骨赘形成、软骨下骨裸露,判断造模成功。造模后模型组大鼠脊髓及中脑的β-EP及CCK-8含量比空白组显著增加(P<0.01),在脊髓层面针刀组及电针组β-EP含量比空白组显著增加(P<0.01),针刀组与模型组之间无显著性差异(P>0.05),电针组与模型组之间有显著性差异(P<0.05),两组的CCK-8含量比模型组显著降低(P<0.01),两组间无显著性差异(P>0.05)。在中脑层面针刀组、电针组β-EP及CCK-8含量比模型组显著降低(P<0.01,P<0.05),两组间无显著性差异(P>0.05)。结论:针刀和电针的刺激可增强β-EP在脊髓水平的释放,加强痛觉信号初级整合中枢的作用。脊髓层面CCK-8的含量并没有随β-EP的升高而升高,这一现象的产生是否由于针刀和电针可对CCK-8的释放产生抑制需要进一步的展开研究。
【Abstract】 Objective:to compare and observe the effects of acupotomy lysis and electro-acupuncture on the neuropeptide corresponding to central pain in rats with knee osteoarthritis Method:divide 80 healthy SD rats randomly into normal control group,model group, acupotomy lysis(AL) group,and electro-acupuncture(EA) group.Mix 4%papain solution with 0.3mol/L cysteine solution in the ratio of 1:1.Pause for 0.5h,inject the mixture,20μL each time,into the left knee joint cavities of rats in model,AL,EA groups on days 1,4, 7.After 4 weeks treat AL group with acupotomy lysis and EA group with electro-acupuncture.Three weeks after treatment,take samples of spinal cord and midbrain from the swellings of rats waists. Measure separately the content of ofβ—EP and CCK-8 therein. Result:cartilage in the affected knee joints of rats are dark yellow.Some are found with eburnation,formation of osteophyte,and exposed subchondral bone.It is therefore determined a successful modeling.There is a conspicuous rise in the content ofβ-EP and CCK-8 in spinal cord and midbrain of model group rats,compared with that of normal control group(P<0.01).With respect to spinal cordβ3 -EP content,AL group and EA group have risen markedly compared with normal control group(P<0.01),no significant difference is found between AL group and model group(P>0.05), and EA group and model group are found to be notably different(P<0.05).With respect to spinal cord CCK-8 content,there is a sharp drop in EA group and AL group compared with model group (P<0.01),and no significant difference is found between EA group and AL group(P>0.05).With respect to midbrainβ-EP and CCK-8 content, AL group and EA group have reduced sharply compared with model group(P<0.01),and no significant difference is found between AL group and EA group. Conclusion:acupotomy lysis and electro-acupuncture give stimulation that boostsβ-EP release in spinal cord,strengthening the function of algesthesia signal preliminary integration centrum.Spinal cord CCK-8 content did not rise withβ-EP.It requires further study to determine if it was because acupotomy lysis and electro-acupuncture restrained CCK-8 release.
