养肝益水颗粒对自发性高血压大鼠血浆AngⅡ、TGFβ1、CTGF含量和肾皮质AngⅡ、TGFβ1、CTGF mRNA表达的影响

【摘要】 目的观察养肝益水颗粒对12周龄自发性高血压大鼠(SHR)血浆AngⅡ、TGFβ1、CTGF含量和肾皮质AngⅡ、TGFβ1、CTGF mRNA表达的影响,以进一步探讨养肝益水颗粒发挥肾脏保护的可能作用机制。方法将40只12周雄性SHR大鼠随机分为模型组、贝拉普利组[1.8mg/(kg·d)]、养肝益水颗粒低剂量组[5.4 g/(kg·d)]、养肝益水颗粒高剂量组[27 g/(kg·d)],每组10只;并设Wistar-Kyoto大鼠(WKY)10只作正常对照组;模型组和正常对照组给予等量生理盐水灌胃,每天1次,共6周。检测各组大鼠血浆AngⅡ、TGFβ1及CTGF浓度和肾皮质AngⅡ、TGF-β1及CTGF mRNA的表达。结果养肝益水颗粒在降低血浆AngⅡ、TGFβ1、CTGF含量的作用上不如贝拉普利,且高、低两种剂量相比无明显差别;在降低肾皮质AngⅡ、TGF-β1及CTGF mRNA表达上,低剂量作用和贝拉普利相似,而高剂量作用超过贝拉普利和低剂量。结论养肝益水颗粒主要通过降低肾脏局部AngⅡ、TGF-β1及CTGF的表达而发挥肾脏保护作用。

【Abstract】 Objective To study the effect of YangGanYiShui Granule(YGYSG) in 12-week-old spontaneously hypertensive rats(SHR) on plasma AngiotensinⅡ(AngⅡ),TGFβ1,CTGF content and renal cortical AngⅡ,TGFβ1,CTGF mRNA expression,and explore the mechanism of YGYSG in the kidney protective.Methods Fifty 12-week-old male SHR were randomized into model group,Bella Plymouth group,YGYSG low dose group,YGYSG high dose group.Other ten Wistar-Kyoto(WKY) were included as control group.Saline were gavaged in model group and control group for six weeks,once a day.Plasma AngⅡ,TGFβ1 and CTGF concentration and renal cortical AngⅡ,TGF-β1,CTGF mRNA expression were measured in each group.Results Bella Plymouth was superior to YGYSG in reducing plasma AngⅡ,TGFβ1 and CTGF,and the high and low dose YGYSG showed no significant difference;in reducing renal AngⅡ,TGF-β1 and CTGF mRNA expression,low doses YGYSG was similar to the role and Bella Plymouth,and the role of high dose YGYSG was superior to low dose YGYSG and Bella Plymouth.Conclusion YGYSG play a role in kidney protection mainly through reducing the kidney local AngⅡ,TGF-β1 and CTGF expression.
