UVA照射对体外培养的皮肤成纤维细胞c-fos,c-jun蛋白表达的影响
Fffect of UVA on c-los and c-jun expression in human skin fibroblasts in vitro

【摘要】 目的:探讨不同剂量UVA对体外培养的人皮肤成纤维细胞c-fos,c-jun蛋白表达的影响,探讨其光老化的作用机制。方法:用组织块培养法进行人皮肤成纤维细胞培养传代,用第5代成纤维细胞做实验对象,用不同剂量UVA(0 J/cm2、UVA 5 J/cm210 J/cm2、UVA 20 J/cm2)照射,倒置相差显微镜下观察细胞形态,MTT法检测细胞增殖活性,细胞免疫组化方法检测c-fos,c-jun蛋白表达。结果:随着UVA照射剂量增加,细胞形态改变明显,部分细胞变圆,细胞增殖活性明显下降,UVA 10 J/cm2、UVA 20 J/cm2照射组OD值下降明显,差异有统计学意义(P<0.05)。随着UVA照射剂量增加,成纤维细胞内c-fos,c-jun蛋白表达增加,10 J/cm2、UVA 20 J/cm2照射组差异有统计学意义(P<0.05)。结论:UVA照射体外培养皮肤成纤维细胞,降低了细胞增殖活性,细胞逐渐变圆,同时使得细胞内AP-1蛋白(c-fos、c-jun)的表达增加降低,导致细胞光损伤,激活了光老化的进程。
【Abstract】 Objictive To investigate effect of UVA on the expression of c-fos and c-jun in human skin fibroblasts in vitro and discuss the mechanism of skin aging.Method Cultured the five generation human skin fibroblast for experiment in tissue culture method and treated with different doses of UVA.Cellular activity was detected by MTT assay.Cell porphology was observed.under light microscope.The expression of c -fos and c -jun were detected by immunocytochemistry.Results Compared with the control group,the decrease of cell proliferation was observed fllowed the dosages-increased of UVA(p<0.05). And parts of cells change from spindle to round,the gap between cells increased.Immunocytochemical result showed that:with the UVA radiation dose increased,c-fos,c-jun were positive in the cytoplasm and nucleus.The expression of c-fos and c-jun was significantly increased(P<0.05). Conclusion UVA treatment skin fibroblast lead to photoaging may according to decrease the proliferation activity,cell injury and increasr the expression of c-fos and c-jun.The activation of AP-1 is important.
