柴胡疏肝散及其拆方对慢性应激抑郁模型大鼠海马组织ERK1/2mRNA表达的影响

【摘要】 目的探讨柴胡疏肝散与其拆方的抗抑郁作用及机制。方法将SD大鼠随机分为正常对照组、模型对照组、全方干预组、拆方干预Ⅰ、Ⅱ组、氟西汀对照组,采用慢性轻度不可预见性应激配合孤养复制抑郁模型。运用敞箱试验和蔗糖水消耗实验观察大鼠行为改变,荧光实时定量PCR(FQ-PCR)检测各组大鼠海马组织细胞外调节蛋白激酶(extracellular regulated protein kinases,ERK1/2)mRNA表达水平。结果与正常对照组比较,模型组大鼠海马组织ERK1/2 2-△△Ct显著降低,差异具有统计学意义(P<0.01),表明模型组海马ERK1/2 mRNA表达明显下调。与模型组比较,全方组、氟西汀组大鼠海马组织ERK1/2 2-△△Ct均显著升高,差异具有统计学意义(P<0.05或P<0.01),拆方Ⅰ组ERK1 2-△△Ct亦明显上调,差异具有统计学意义(P<0.05);而拆方Ⅱ组海马组织ERK1/2 2-△Ct均无明显增高,与模型组比较差异无统计学意义(P>0.05)。结论慢性轻度不可预见性刺激结合孤养可诱导大鼠脑组织ERK1/2 mRNA表达下调,柴胡疏肝散可能通过促进海马组织ERK1/2 mRNA的表达,改善抑郁大鼠模型的行为学异常,发挥抗抑郁作用;拆方Ⅰ组也具有较好的抗抑郁作用,但作用不及全方组;拆方Ⅱ组则无明显抗抑郁作用,但具增强抗抑郁作用。

【Abstract】 Objective To explore the antidepressant effects and mechanism of Chaihu Shugan San composition,a compound traditional Chinese herb medicine,and its components.Methods Rats were randomly divided into six groups:Normal control group,depression model group,CHSGS group,componentⅠ,Ⅱgroups and Fluoxetine group.The depression model was replicated by chronic unpredictable mild stress and single house for 28 days.Behavioral scores of rats were detected by open-field test and sucrose water consumption test,and ERK1/2 mRNA expression in Hippocampus tissue was assayed by Real-time fluorescent quantitative PCR.Results ERK1/2 mRNA expression was down-regulated in the depression model group compared with the normal control group(P<0.01).Compared with the model group,ERK1/2 mRNA expression in CHSGS group and Fluoxetine group was both up-regulated,and only ERK1 mRNA expression in componentⅠgroup was up-regulated(P<0.05or P<0.01 );No significant difference is exist between componentⅡgroup and model group(P>0.05).Conclusion Isolated-living condition and chronic mild unpredictable stress can down-regulate the expression of ERK1/2 mRNA in Hippocampus tissue. CHSGS might exert an antidepressant effect through decreasing expression of ERK1/2 mRNA in hippocampus,its componentⅠmight play an important role in its anti-depression effect,while compatibility of the use of the herbs componentⅡcould enhance antidepressant efficacy.
