逍遥散对肝郁脾虚证模型大鼠海马和杏仁核AMPA受体基因表达的调节机制
RT-PCR results reveal the possibility of effective regulation channel of Xiaoyao san(XYS)treating rats with Liver stagnation and Spleen deficiency syndrome(LSSDS)

【摘要】 目的:前期实验利用慢性束缚方法成功复制出大鼠肝郁脾虚证模型。本实验在此模型基础上,运用脑立体定位仪于双侧杏仁核微量注射CNQX,CNQX拮抗杏仁核AMPA受体,可以缓解海马CA1、CA3神经元受损,起到保护海马神经元作用。同CNQX作用机制进行对比,试图探讨逍遥散治疗肝郁脾虚证的一条重要调节通路。方法:75只SD大鼠随机等分为5组,正常组、模型组、假手术组、CNQX组和逍遥散组。分别以前三组作为参照,排除混杂因子,比较CNQX组和逍遥散组分别在海马CA1、CA3和杏仁核如下指标GluR1 mRNA、GluR2 mRNA表达变化趋势是否一致。结果:(1)海马CA1和CA3区:①GluR1 mRNA表达:在CA1区,与正常组比较,模型组和假手术组均有统计学意义(均P<0.01),且均较正常组增高;在CA3,与正常组比较,模型组和假手术组均有统计学意义(均P<0.01),且均较正常组GluR1 mRNA表达减少;在CA1和CA3区,正常组、CNQX组和逍遥散组间均无统计学意义(均P>0.05)。②GluR2 mRNA表达:在海马CA1、CA3区,与正常组比较,模型组和假手术组均有统计学意义,且均较正常组减少;正常组、CNQX组和逍遥散组间无统计学意义(均P>0.05)。(2)杏仁核BLA区:CNQX组于拮抗AMPA受体,GluR1 mRNA和GluR2 mRNA均表达最低;其它各组这二指标变化趋势与CA1区各组间变化趋势均分别相反,且正常组和逍遥散纽间均无统计学意义(均P>0.05)。结论:以假手术组或模型组为参照,逍遥散组和CNQX组上述二指标RT-PCR表达结果在海马和杏仁核变化趋势均相似,推断逍遥散可能有一调节通路与CNQX调节机制吻合,即可能通过抑制杏仁核,尤其抑制其BLA区兴奋,缓解海马各区受损,起到调节作用。纠正杏仁核和海马的"兴奋-抑制"失衡,重建稳态,可能是逍遥散治疗肝郁脾虚证的一条重要调节通路。
【Abstract】 Objective:With good operability and repeatability,our group ensure the rat model subjected to chronic immobilization stress(CIS)was an ideal model for research on LSSDS.This experiment was designed to explore CIS rats’ RT-PCR results in hippocampal sub-regions CA1,CA3 and amygdaloid sub-region BLA after suppressing both amygdaloid AMPARs by microinjection of CNQX. Compare XYS group with CNQX group,analysis of these findings may reveal the possibility of molecular mechanisms and effective regulation channel of XYS treating this syndrome.Methods:75 male SD rats were randomly divided into 5 equal groups:normal group,CIS group,sham-operation group, CNQX group and XYS group.GluR1,2 mRNA in hippocampal sub-regions CA1,CA3 and amygdaloid sub-region BLA in these 5 groups were examined by RT-PCR test.Results:the expressional tendency of AMPA receptors subunits GluR1 mRNA and GluR2 mRNA were different in hippocampus;so did the region amygdala.respectively compared with model or sham operation group,the expressional tendency of AMPA receptors subunits was almost the same in CNQX and XYS group.This finding revealed that XYS may act the same as CNQX,thus XYS afforded effective protection against LSSDS through suppression of both amygdaloid excitability.Conclusion:the balance coordination between hippocampus and amygdala may be the key in transformation from adaption to damage in a state of stress,is one of the central mechanisms of Liver governs smoothing qi flow(LGSQF) on regulating the stress.XYS may effectively regulate the balance of the excitability of hippocampus and amygdala.This hypothesis may reveal the molecular mechanisms and regulation channel on XYS affording effective protection against Liver invading the spleen syndrome.
