保鲜剂处理养殖大黄鱼冷藏过程优势腐败菌动态变化研究

【摘要】 研究了冷藏(4±0.5℃)过程中对照组和生物保鲜剂处理组(A:0.2%茶多酚+2%壳聚糖;B:0.2%迷迭香+2%壳聚糖)养殖大黄鱼细菌种类和数量的变化。试验采用特定的选择性培养基分离培养了冷藏过程中养殖大黄鱼的细菌总数、酵母菌属、假单胞菌属、乳酸菌属、微球菌属、明亮发光杆菌和腐败希瓦氏菌属等7类微生物,同时采用PCR-DGGE技术与传统选择性培养基的方法进行了对比试验。研究结果表明:对照组和处理组的优势腐败菌主要为腐败希瓦氏菌,其次为假单胞菌和明亮发光杆菌,该结果与DGGE(变性梯度凝胶电泳)电泳图谱表现结果相似;同时经过生物保鲜剂处理组的腐败希瓦氏菌、假单胞菌和明亮发光杆菌在冷藏过程中受到了明显的抑制,尤其对腐败希瓦氏菌抑制效果最为明显。根据养殖大黄鱼的鲜度评定标准,冷藏条件下,A和B处理组有非常明显的抑菌效果,其货架期延长了8-10天。
【Abstract】 Types and amounts changes of bacterial flora in large yellow croaker during refrigeration(4±0.5℃)were studied,including control and treatment groups(A:0.2%TP+2%chitosan,B:0.2%rosemary + 2%chitosan).The total viable counts,Saccharomyces,Pesudomonas,Lactic acid bacteria,Micrococcus, Photobacterium phosphoreum,Shewanella putrefaciens were analysed in the experiment by applying the traditional method of selective media as well as PCR-DGGE technology.The microbiological analysis results showed that dominated spoilage organisms of the large yellow fish was Shewanella putrefaciens,and the secondary prepotent floras were Pesudomona and Photobacterium phosphoreum.The results were similar to DGGE(denaturing gradient gel electrophoresis)electrophoresis.By biological preservatives treatment, the Shewanella putrefaciens,Pesudomona and Photobacterium phosphoreum were inhibited during refrigerated storage,especially the amount of Shewanella putrefaciens decreased obviously.According to the fresh assessment standards of the large yellow croaker,the bacterial flora of A and B treatment groups were inhibited significantly,and the shelf life of large yellow croaker were extended 8-10 days.
