槲皮素在小鼠血浆中代谢产物分析与检测

【摘要】 本研究旨在建立简便分离小鼠血浆中槲皮素脂溶性与水溶性代谢产物的方法,并建立槲皮素代谢产物的O-TOF(四级杆飞行时间质谱)检测方法。实验中采用三周龄小鼠灌胃槲皮素4h后眼眶取血,血浆经过LH-20柱正己烷/氯仿、甲醇洗脱分离成正己烷相和甲醇相,同时对得到的正己烷洗脱相加入胰脂肪酶酶解,甲醇洗脱相加入β-葡萄糖醛酸酶、硫酸酯酶酶解,采用Q-TOF法对槲皮素代谢产物进行定性定量检测分析。通过与标准品比较,在分析检测条件下可稳定检测到血浆样品中的槲皮素与异鼠李素。同时血浆的水溶性分离相能检测到槲皮素和异鼠李素,其中异鼠李素结合酯居多;血浆的脂溶性分离相能检测到槲皮素和异鼠李素,且槲皮素呈现较高峰,异鼠李素峰较弱。槲皮素在血液中原形代谢脂溶性代谢产物占83.79%,水溶性代谢产物较少,槲皮素与脂肪酸成酯在槲皮素代谢中占主要部分。该方法准确性高、重现性好,可有效分离和检测血液中槲皮素脂溶性和水溶性代谢产物。

【Abstract】 This stuely aimed at Establishing a simple method to separate quercetin’s lipo-soluble and water -soluble metabolites in mice’s plasma,and setting up a method to detect quercetin’s metabolites with Q-TOF(Quadrupole-time of flight mass spectrometry).In the stndy,3 weeks-old Kunming mice are administrated quercetin 100 mg·Kg-1 by gavaged and blood are collected after 4h.Quercetin’s lipo-soluble and water-soluble metabolites in plasma are firstly separated by N-hexane/chloroform(1:1 )and methanol with LH-20,furthermore N-hexane elution part is hydrolyzed by pancreatic lipase enzyme,while methanol elution part is hydrolyzed by sulfatase andβ-glucuronidase.Then analysed by Q-TOF scanning in negative ion conditions.Compared with the standard,quercetin,isorhamnetin of plasma sample are stability detected in the analysis condition.Quercetin and isorhamnetin can be detected in water-soluble phase,and isorhamnetin ester takes up a majority proportion.The lipo-soluble phase can also detect the quercetin and isorhamnetin, while quercetin presents a high peak,isorhamnetin presents a weaker.Quercetin’s lipo-soluble metabolites part accounts for 83.79%in the plasma,while the water-soluble part only takes up a small proportion. The reported method has high accuracy and reproducibility;therefore,it can separate and detect quercetin’s liposoluble and water-soluble metabolites in mice’s plasma effectively.
