不同剂量矽尘染毒对实验大鼠体内SP-D含量的影响

【摘要】 目的探讨不同剂量矽尘对大鼠血清和BALF中SP-D表达水平的影响。方法选用健康成年清洁级雄性SD大鼠32只,体重180-200g,按体重随机分成4组,每组8只。对照组注入无菌生理盐水1ml;染矽尘组:分别注入12.5mg/ml(低剂量组)、25mg/ml(中剂量组)和50mg/ml(高剂量组)二氧化硅混悬液1ml。大鼠在屏障动物实验室饲养,自由饮水。染毒4周后,处死动物,心脏取血分离血清;采用支气管肺泡灌洗(Bronchoalveolar lavage,BAL)方法收集支气管肺泡灌洗液(BALF),离心后收集上清液,用考马斯亮蓝法测定BALF中总蛋白含量,同时测定BALF中总抗氧化能力(T-AOC)、羟脯氨酸(HYP)、丙二醛(MDA)的含量;用ELISA法分别测定BALF和血清中的SP-D的含量;取肺组织做HE染色,观察肺组织病理变化。结果 BALF中总蛋白、HYP和MDA含量随着染矽尘剂量的升高而升高;低剂量组T-AOC显著高于对照组,而中、高剂量组T-AOC显著低于对照组;染矽尘组大鼠血清和BALF中SP-D含量显著低于对照组,但各染毒组间差别不大;病理学检查结果显示对照组大鼠肺泡结构清晰、肺泡壁薄、血管周围无明显细胞浸润,染矽尘组大鼠肺泡内有有不同程度的巨噬细胞和中性粒细胞浸润,肺泡间隔增厚、水肿,高剂量组大鼠肺泡出现尘细胞结节。结论 SP-D水平变化可能在矽尘所致的肺纤维化中发挥作用。
【Abstract】 Objective To investigate the effect of different doses of silicon on the expression of SP-D in rats. Methods 32 SD rats,weighting 180-200g,were randomly divided into control group,12.5mg/ml,25mg/ml and 50mg/ml silicon groups.Rats in silicon groups were infused with 1ml different doses of quartz dust suspension into tracheas to establish the silicosis mode.The control group was treated with 1ml sterilized saline in the same way.On 28d after establishment of the animal model,rats were sacrificed and lung tissues were taken.Total protein,HYP,MDA concentration and T-AOC in BALF were measured.The levels of SP-D in serum and BALF were determined by ELISA.Lung tissue were stained by hematoxylin and eosin,and observed by a microscope. Results HE staining proved that the rat model of silicosis was successfully established.Compared with the control group,the levels of Total protein,HYP,MDA in BALF were higher in silicon groups rats.Meanwhile the levels of T-AOC in 12.5mg/ml silicon group were higher than in the control group,but were decreased in 25mg/ml and 50mg/ml silicon groups.The levels of SP-D in serum and BALF were decreased in silicon groups rats and the differences were significant.Conclusion Decreased expression of SP-D in serum and BALF might involved in the process of lung fibrosis induced by silica dust.
