茶树离体增殖与再生体系的建立

【摘要】 试验以龙井43茶树腋芽离体培养获得的无菌植株作为研究材料,探讨适宜继代增殖、再生和生根的条件。试验结果表明:龙井-43的最佳增殖条件是以1/2MS为基本培养基,添加0.1μmol/L的TDZ,配合使用0.49μmol/L IBA,增殖倍数可以达到2.0以上,茶苗增殖倍数随着TDZ浓度的升高而降低,在含有TDZ的培养基中连续继代也不利于茶苗的增殖与伸长,茶苗在不添加任何植物生长调节剂培养基中能够良好地伸育;茶苗茎段基部TDZ0.1μmol/L+0.49μmol/L IBA培养基上诱导的愈伤组织,继代转入BA 8.88μmol/L+IBA0.49μmol/L培养基中,可以诱导出不定芽;使用2450μmol/L IBA浸泡茶苗基部5min,诱导生根效果明显。诱导2周后就开始有茶苗生根,第6周生根率达到70.8%,茶苗平均根数3.8条,茶苗炼苗后,移栽至灭菌的基质中(营养土:蛭石=1:1),移栽后30d,茶苗成活率为63.6%。

【Abstract】 Shoot multiplication system and plantlet regeneration system were investigated. Maximum number of shoots was developed on 1/2 MS medium containing 0.1μmol/L Thidiazuron (TDZ) and 0.49μmol/L IBA.Number of shoots formed in explants decreased when successive subculture on medium containing 0.1μmol/L TDZ and failed to develop into shoots.Shoot elongation was achieved when shoots were cultured on hormone-free 1/2 MS medium.The shoots produced compact,nodular callus from the basal of shoots on 1/2 MS meidium containing 0.1μmol/L TDZ and 0.49μmol/L IBA,when shoots connected with callus were transferred to 1/2 MS medium supplemented with 8.88μmol/L BA,0.49μmol/L IBA,the callus germinated into plantlets. For rooting,tea shoots were treated with 2450μmol/L IBA solution for 5 minutes and transferred directly to hormone-free 1/4 MS medium.After 6 weeks,70.8%of them produced roots,mean number of root per micro-shoots was 3.8.The rooted plants were transferred to a sterile mixture of peat and soil(1:1) in bottles,irrigated with sterile water.After 30 days,the frequency of survival was 63.6%.
