【摘要】 目的:建立测定肉苁蓉及管花肉苁蓉中半乳糖醇的含量测定方法,为中药肉苁蓉标准的制定提供依据。方法:通过建立半乳糖醇的HPLC含量测定方法,对20个批次的肉苁蓉中半乳糖醇进行分析。色谱条件为Alltima C₁₈色谱柱（4.6mm×250mm,5μm）;流动相为乙腈-水（体积比80:20）溶液。流速为1.0mL·min^-1;进样量为10μL;ELSD检测器漂流管温度为80℃,载气流速为3.0L·min^-1。结果:半乳糖醇89.6～1120mg·L^-1范围内峰面积对数值与进样量对数值有良好的线性关系（r=0.9990）,方法检出限（MDL=3.14 SD）为15.998mg·L^-1,定量限为79.903mg·L^-1,肉苁蓉和管花肉苁蓉中半乳糖醇的平均回收率（n=5）分别为100.34%和101.11%,RSD分别为1.72%和1.33%。结论该方法简便准确,灵敏度高、重复性好,可以作为肉苁蓉的质量控制方法。更多还原

【Abstract】 OBJECTIVE:To establish an HPLC method for the determination of galactitol in Cistanche deserticola and Cistanche tubulosa and to provide evidence for the quality control of Cistanches Herba.METHODS:The content of galactitol in 20 batches of Cistanches Herba samples were determined using the established HPLC method.The samples were separated by an Alltima C₁₈ column（4.6 mm×250 mm,5μm） with acetonitrile-water（80:20 by volume ratio） as mobile phase.The flow rate was 1.0 mL·min^-1 and the injection volume was 10μL.Shift tube temperature of ELSD was set at 80℃and the gas flow rate was 3.0 L·min^-1.RESULTS:The linear range of galactitol was 89.6～1120 mg·L^-1（r = 0.9990）,Method Detection Limit was 15.998 mg·L^-1,Limit of Quantitation was 79.903 mg·L^-1.The average recovery rate of galactitol in Cistanche deserticola and Cistanche tubulosa was 100.34%and 101.11%respectively with a RSD of 1.72%and 1.33%（n = 5）.CONCLUSION:The method is sensitive,repeatable and accurate, which can be used for the quality control of Cistanches Herba.更多还原
