内部胶凝化在脂质体冷冻干燥中的稳定作用

【摘要】 目的制备载紫杉醇内部胶凝化脂质体,考察内部胶凝在冷冻干燥中对脂质体的稳定作用。方法搅拌子法测定明胶体系胶凝温度;薄膜分散法制备载紫杉醇内部胶凝化脂质体;与普通脂质体及含支架剂脂质体比较,考察冻干前后内部胶化对脂质体包封率和粒径的影响。结果随浓度增大明胶体系的胶凝温度升高,内部胶凝化脂质体的包封率不受明胶体系的影响,均在82%以上,冻干再分散后内部胶凝化脂质体包封率维持在84%以上,而普通及含支架剂脂质体均降至20%左右。结论内部胶凝化在脂质体冷冻干燥过程中可起到稳定作用。

【Abstract】 OBJECTIVE To study the influence of interior gelatinization on the stability of freeze dried liposomes with paclitaxel loaded. METHODS The sol-gel transition temperatures (Tgel) of gelatin solutions were determined by stirring method. A film dispersion method was adopted to prepare the pacliaxel loaded interior gelatinized liposomes (PGLs). And the effects of freeze drying on entrapment efficiency and particle sizes were compared among conventional liposomes, liposomes containing scaffolds and PGLs. RESULTS Tgel was increased with increasing of gelatin concentration. The entrapment efficiencies of the PGLs all exceeded 82% regardless of the gelatin concentrations. Freeze-dried PGLs were easily reconstituted, and the entrapment efficiency was maintained above 84%. However, entrapment efficiency of conventional liposomes and that containing scaffolds were all decreased to about 20%. CONCLUSION The interior gelatinization would stabilize liposomes in freeze drying.
