丹参水溶性成分的纯化工艺和质量研究

【摘要】 目的:提取纯化丹参中水溶性酚酸类有效成分,并进行相应的质量分析研究。方法:以HPLC检测丹酚酸B的含量,采用单因素和正交实验优化丹参的提取工艺;采用大孔吸附树脂方法纯化丹参水溶性成分;并对丹参及其提取物的水溶性成分进行HPLC指纹图谱研究,制定相应质量标准。结果:丹参中主要水溶性成分丹酚酸B的提取转移率达85%,提取纯化物中含丹酚酸B大于65%、丹参素8%,其他丹酚酸12%。相应HPLC指纹图谱分析方法可有效分析和鉴别主要水溶性特征峰。结论:本方法能够快速高效分离纯化丹参水溶性成/组分,得到较高纯度的丹参总酚酸;HPLC指纹图谱分析方法的精密度、重现性良好。

【Abstract】 Objective:To extract and purify the active components of salvia miltiorrhiza, and to determinate the relevant quality analysis. Methods:The HPLC method of salvianolic acid B was established. The factors influencing the extraction were invested with a single-factor and orthogonal design. The water-soluble components of salvia miltiorrhiza were purified with macroporous resin. The HPLC fingerprint of Salvia miltiorrhiza and its purified extract were study, and the quality specification was established. Results:The transfer rate of salvianolic acid B was up to 85%, and the contents of salvianolic acid B and tanshinol were respectively 65% and 8%, and other salvianolic acid about 12%. The HPLC fingerprint analysis method can effectively analyze and identify the main characteristic peaks of water-soluble components. Conclusion:This method can quickly and efficiently purified Salvia miltiorrhiza water-soluble components. The salvianolic acid components that had higher purity were obtained. The HPLC fingerprint method has good precision and reproducibility.
