紫苏子油的化学成分研究

【摘要】 目的:对超临界CO2萃取的紫苏子油的化学成分进行研究。方法:采用超临界二氧化碳流体萃取法(SFE-CO2)提取;通过气相色谱-质谱联用技术(GC-MS)分析鉴定化合物结构,色谱条件:石英毛细管柱HP-Innowax(30m×0.25mm×0.50μm),进样口温度250℃,程序升温,柱温8O℃,保持5min,以5℃·min-1的速度升至120℃,保持10mim,再以6℃·min-1升温至230℃,保持30min。载气He,进样量为1微升,分流比1:5。EI源温度230℃,电子轰击能量为7OeV,全离子扫描范围m/z40~500,以峰面积归一化法计算各成分相对百分含量。结果:分析鉴定了79种化学成分,所鉴定的成分占总流出峰面积的91.59%。结论:为紫苏子油的药效物质基础进一步研究提供科学依据。

【Abstract】 Objective:Analyze the constituents in supercritical fluid CO2 extraction(SFE-CO2)of perilla seed oil with gas chromatography mass spectrometry.Methods:Perilla seed oil was extracted with SFE-CO2,and structure analyzed by GC-MS.The GC-MS analysis with a quartz capillary column HP-Innowax (30m×250um×0.50um film thickness)was used.The inlet temperature was maintained at 250℃.The column oven was begun at 80℃,held for 5 min.Then programmed from 80 to 120℃ at 5℃ ·min-1,held for 10 min, programmed 230 ℃ at 6 ℃· min-1,held for 30 min.Helium was used as the carrier gas.The inlet volum was 1 μl, diffluence rate was 1:5. The mass spectrometry conditions were as follows:ionization energy,70eV;ion source temperature,230℃.The mass selective detectorwas operated in the TIC mode(m/z 40-500).Results:By the analysis of the SFE-CO2 extractions of perilla seed oil 79 peaks were separated and identified,the compounds were quantitatively determined by normalization method.The identified compounds represent 91.59% of total GC peak areas.Conclusion:The results will provide important foundation for understanding the constituents and further exploitation of perilla seed oil.
