阴道给药后纳米银凝胶在动物体内的分布和蓄积特性

【摘要】 研究纳米银凝胶在兔体内的吸收和分布情况,以了解纳米银凝胶中的纳米银是否具有在体内迁移和蓄积的特性。将成年新西兰雌兔经阴道注入纳米银凝胶,采用5倍临床给药剂量(0.58g·kg-1·d-1),连续给药6天后,在预定时间点解剖动物并取肝、肾、脾、子宫、阴道等主要组织器官,利用超高压微波消解系统消解并用原子吸收光谱仪检测Ag含量。结果显示,给药组在给药6天后的8小时,纳米银在各组织器官中的银含量达到高峰,与对照组各脏器中银含量相比有显著性差异(P<0.05)。其中阴道是纳米银主要的分布器官,其银含量高于对照组达20倍以上。1周以后各组织中的银含量均大幅度下降,到1个月和3个月后,各试验组脏器中的银含量与对照组无显著性差异(P>0.05)。以上结果表明纳米银可以通过血液循环系统迁移,并在体内肝、肾、脾、子宫、阴道等主要脏器中分布,其中经阴道粘膜的局部吸收量较高。给药结束后1个月,纳米银凝胶在这些脏器中无明显蓄积。纳米银凝胶在体内的存在状态、进入血液系统的方式和对各部位组织细胞的影响,尤其是阴道粘膜局部的病理变化等需要进行进一步的系统研究。

【Abstract】 To investigate the absorption,distribution and accumulation of nano-silver gel in the rabbit,and to analyze whether the nano-silver gel have the character of translocation and accumulation,adult female New Zealand rabbits were adopted with nano-silver gel at 5 times the clinical dose(0.58 g·kg-1·d-1) via vagina,and then they were sacrificed at the time points after 6 days continuous administration.The main organs of the experimental animals were harvested for silver content analysis by using the extra high pressure microwave and the atom absorption spectrum instrument.Eight hours after the last dose administration,the nano-silver content were remarkably increase compared to the control groups(P<0.05).The nano-silver content in vagina was significantly high,in which the silver content was 20 times more than in control groups.Thesilver content in each organ significantly decreased after a week post-treatment,and had not remarkable difference in each organ after 1 month and 3 months post-treatment compared to the control groups(P>0.05).The nano-silver gel was absorbed by vagina,can translocated and distributed in liver,kidney,spleen and womb etc.through blood system.The nano-silver gel has not shown obviously accumulation in these organs after one month post-treatment.The further study are needed for investigating the properties of nano-silver gel in the body,the pathway of nano-silver were taken-up into blood system,and the toxicity to cells or tissues.
