西藏小型猪细胞色素b的分子遗传学研究

【摘要】 目的本研究旨在对西藏小型猪Cyt b基因序列和氨基酸序列进行分析,研究其遗传分化及其与欧洲品种猪和国内几种地方猪的亲缘关系。结果表明西藏小型猪Cyt b基因序列有9个变异位点,核苷酸多样性为0.0789。国内猪包括西藏小型猪的Cyt b基因序列与欧洲品种猪相比有16个主要突变位点,其中西藏小型猪有2个特殊碱基位点:420位点的T→C转换和883位点的G→A转换,几乎各占50%。氨基酸序列分析表明,西藏小型猪与欧洲品种猪有3个氨基酸位点存在着差异,其中在295位点,一部分西藏小型猪与以上其它几种品种猪同为缬氨酸(Ⅴ),而另一部分西藏小型猪为异亮氨酸(Ⅰ)。结果显示只有部分西藏小型猪与巴马小型猪、贵州香猪、五指山猪有很近的亲缘关系,同时证实西藏小型猪群体内存在遗传分化。

【Abstract】 Objective To analyze the genetic differentiation of Tibet mini-pigs and relationships with other pigs including Europe and domestic pigs,the sequences of Cyt b gene and amino acid were sequenced.The results showed that there were 9 mutation sites in Cyt b gene of Tibet mini-pigs and the nucleotide diversity was 0.789.There were 16 mutations between domestic pigs in China and pigs from Europe.There was significant difference in two nucleotide sites:a T-C switch at site 420 and a G-A switch at site 883,with a proportion of 50%,respectively.The amino acid sequence analysis indicated that Tibet mini-pigs,compared with pigs from Europe,were different at three amino acid sites,especially at site 295,a part of Tibet mini-pig together with other pigs,Valine,while another,Isoleucine at this site.Therefore,the partial of Tibet mini-pigs had very closely blood relationships with Bama miniature pigs,Guizhou xiang pigs and WZS pigs,and confirmed that there was genetic differentiation in Tibet mini-pig group.
