Runx2对成釉细胞分化和釉质发育的作用

【摘要】 目的:研究Runx2对成釉细胞分化的影响和对釉质发育的作用。方法:构建在成釉细胞中特异性表达Runx2的转基因小鼠,用qRT-PCR方法检测出生后3日小鼠的釉原蛋白表达量。进行牙齿发育不同阶段的组织学检测。利用Micro-X ray,Micro-CT,扫描电镜等技术检测转基因小鼠釉质发育的变化。结果:转基因小鼠的成釉细胞失去原有的高柱状形态和极化特征。釉原蛋白表达显著下调。釉基质的的分泌在分泌的早期阶段就已完全阻断,最终造成釉质缺失。结论:Runx2抑制了釉原蛋白的表达和成釉细胞的分化,造成釉基质形成障碍,阻断了釉质的发育。
【Abstract】 Objectives:To study the role of Runx2 in differenciation of ameloblast and development of enamel.Methods:We generated transgenic mice in which runx2 was expressed specifically in ameloblast.We measured the expression level of amelogenin in 3 day’s tooth germ and did histological analysis of the molar in different development ages. Micro-X ray,Micro-CT and SEM were performed to observe the abnormal changes in the enamel development of the transgenic mice.Results:Ameloblasts lost their polarization and tall columnar shape.The expression of amelogenin was down-regulated obviously.The formation of enamel matrix was completely blocked at the very beginning of the enamel matrix secreting stage,resulting in enamel lost.Conclusion:Runx2 inhibited the expression of amelogenin and ameloblast differentiation,interfered with enamel matrix development and finally stopped enamel formation.
