单束定点辐射线虫咽部的生物学效应初探及线虫固定方法研究

【摘要】 以遗传背景清楚,遗传上可操纵性强,个体微小,整体透明,容易被各种粒子穿透的模式生物线虫作为研究对象,进行辐射损伤信号在个体水平上的转导及其机理研究。为了实现精确定点辐照,需进行活体线虫的固定。验结果表明,乙醚:无水乙醇=1:1混合使用,能使线虫快速麻醉,并使线虫在整个辐照过程中处于麻醉状态,固定不移动,辐照结束后可快速复苏,复苏率达100%。在此基础上,进行定点辐照线虫咽部,检测不同剂量条件下线虫产卵量和尾部性腺细胞凋亡的变化。初步试验结果显示,定点辐照线虫咽部5000partices,使得线虫尾部性腺细胞凋亡增加,达显著性差异。

【Abstract】 Using C.elegans- with clear genetic background,easy genetic maneuverability,small individual,transparence, easily penetrated by a variety of particle as a in vivo model organism,irradiation damage at the individual level of the signal transduction and the mechanism research were investigated.In order to radiate accurately,the worms need Immobilize.The results showed that the ether:ethanol = 1:1 mixture,enabled the worms quickly anesthesia,and kept the worms Immobilization in the whole irradiation process,then quickly recovered after irradiation and recovery rate of 100%.On the basis,the laved and the apoptotic cells in the distal gonad would be test when the worm pharynx were irradiated by single-particle microbeam.The primary results showed that the apoptotic cells in distal gonad significantly increased when the worm pharynx were irradiated 5000particles.
