绵羊β-防御素cDNA的克隆及其真核表达载体构建

【摘要】 绵羊β-防御素(SBD-1)与T载体成功连接,并将其成熟肽编码片段亚克隆到了毕赤酵母表达载体Ppic9k上,构建了真核表达质粒Ppic9k-mSBD-1。且将其在毕赤酵母GS115中进行了分泌表达。将表达产物纯化后进行了活性鉴定。结果表明,mSBD-1对大肠杆菌和金黄色葡萄球菌均有抑制作用,而且对金黄色葡萄球菌的抑制作用更为明显。
【Abstract】 the ovis β-defensin(SBD-1)was linked with the T vector.And the region of coding mature peptide was subcloned with the Ppic9k(Pichia pastoris expression vector). The recombination vector Ppic9k-mSBD-1 was constructed.The mSBD-1 was secreted in GS115. And the secretion was purified and the activity was determined.The result showed that mSBD-1 can inhibit the growth of the Escherichia coli and Staphylococcus aureus,and the inhibition was significantly on Staphylococcus aureus.
