丹蛭降糖胶囊对胰岛素抵抗大鼠大网膜脂肪细胞PPAR-γ mRNA表达的影响

【摘要】 目的:观察丹蛭降糖胶囊对小剂量链脲佐菌素(STZ)加高热量饮食诱导的胰岛素抵抗(IR)模型大鼠大网膜脂肪细胞过氧化物酶体增生物激活受体-γ(PPAR-γ)mRNA表达的影响。方法:雄性Wistar大鼠随机分为空白组、模型组、治疗组(马来酸罗格列酮组、丹蛭降糖胶囊高剂量+马来酸罗格列酮组、丹蛭降糖胶囊低剂量组、丹蛭降糖胶囊高剂量组),正常组喂以普通饲料,模型组与治疗组注射小剂量STZ并喂以高热量饲料,常规测定各组大鼠治疗前后的空腹血糖(FPG)和治疗后各组大鼠的空腹血清胰岛素水平(FIns),计算胰岛素敏感性指数(ISI),提取大网膜脂肪组织总RNA,采用逆转录PCR技术扩增PPAR-γ基因片段,检测其表达水平。结果:丹蛭降糖胶囊能降低模型大鼠FPG水平和FIns含量,提高ISI。与正常组比较,模型组PPAR-γmRNA表达明显减少(P<0.01),模型组与丹蛭降糖胶囊低剂量组PPAR-γmRNA表达量无明显差异(P>0.05),与模型组比较,马来酸罗格列酮组、丹蛭降糖胶囊高剂量+马来酸罗格列酮组、丹蛭降糖胶囊高剂量组PPAR-γmRNA表达显著增加(P<0.01)、丹蛭降糖胶囊高剂量组PPAR-γmRNA表达明显增加(P<0.05)。结论:丹蛭降糖胶囊对大鼠IR有显著的改善作用,提示其机制与中药复方的作用是多途径靶点的,可能与提高IR大鼠脂肪细胞PPAR-γ基因的表达有关。
【Abstract】 Objective:Investigate Danzhi Jiangtang Capsule’s effect on peroxisome proliferator-activated receptor-γ(PPAR-γ)mRNA’s expression in greater omental adipocyte in insulin resistance(IR)rats which were induced by low dose of streptozotocin(STZ)and high calorie food.Methods:Male Wistar rats were randomly divided into six groups(normal group、model group、Rosiglitazone Maleate Tablets treating group、Rosiglitazone Maleate Tablets and high dose of Danzhi Jiangtang Capsule treating group、low dose of Danzhi Jiangtang Capsule treating group、high dose of Danzhi Jiangtang Capsule treating group).The rats of normal group were fed with common food while the others with high calorie food and injected with low dose of STZ.Fasting plasma glucose(FPG)level and fasting insulin(FIns)level of all rats were measured by routine methods,and insulin sensitivity index(ISI)was calculated accordingly.At the end of experimental treatment,the total mRNAs of greater omental adipose tissues were extracted and the genic fragments of PPAR-γwere amplificated in order to detect their expression levels.Results:Danzhi Jiangtang Capsule could reduce the FPG level and FIns level in model rats and enhance ISI effectively.The PPAR-γmRNAs were expressed less obviously in the rats of model group than in normal group(P<0.01).The expressional level is similiar in the rats of low dose of Danzhi Jiangtang Capsule treating group and the model group,but it is significantly higher in the other three treating groups than the model group(P<0.05).Conclusions:Danzhi Jiangtang Capsule can ameliorate IR condition of the model rats and its mechanism is probably related to its effect of enhancing the genic expressional level of PPAR-γ.
