蛛网膜下腔出血后RHO激酶对脑血管痉挛的调控研究

【摘要】 目的探讨RHO激酶在蛛网膜下腔出血后患者脑脊液中的表达及其在脑血管痉挛的调控作用。方法采集35例SAH患者脑脊液标本,采集时间为入院后第1、3、7、10、14天,RT-PCR方法检测Rho激酶mRNA的表达,采用放免法检测脑脊液中内皮素-1浓度,采用TCD检测颅内主要动脉血流速度。结果出血后第3天脑脊液中Rho激酶表达增高,在出血后第7天达到高峰,而后逐渐下降。ET-1浓度于出血后第3天脑脊液中即有明显升高,出血后第7天达到高峰,而后逐渐下降。同时,血管痉挛患者Rho激酶表达明显高于未痉挛者。结论SAH患者脑脊液中Rho激酶的表达与ET-1浓度梯度有相关性,参与SAH后脑血管痉挛的发生,其含量与病情演变、脑血管痉挛CVS程度有关。
【Abstract】 Objective To investigate the dynamical changes of Rho kinase mRNA of the cerebrospinal fluid CSF and its relatienship with cerebral vascular spasm CVS. Methods ET-1 in CSF during the 1 st,3 rd,the 7 th and the 10 th,14th day after subarachn-oid hemorrhage were colected.The amounts of Rho-kinase mRNA in CSF were determin-ed by RT-PCR,TCD was used to measure the velocily of the cerebral artery Results The levels of ET-1 and Rho-kinase mRNA in CSF were remarkably increased onthe 3rd day and reached at the peak-point on the 7th day after subarachn-oid hemorrhage,which were significantly higher than those in those without CVS. Conclusion There is a positive correlation between the level of Rho-kinase mRNA and ET-1 in CSF,they all participate in the change of symptom CVS.
