黄芪多糖对高血压病血管内皮细胞TLR4、NF-?B表达影响

【摘要】 目的:观察黄芪多糖对TLR-NF-?B信号转导通路的影响,探讨其保护高血压病血管内皮细胞损伤的机制。方法:10%的高血压病患者及健康人血清干预人脐静脉内皮细胞(HUVEC-C)24h,实时荧光定量(PCR)法测TLR4 mRNA表达。不同浓度的黄芪多糖干预脂多糖诱导的HUVEC-C TLR4表达(24h)及核转录因子(NF-?B)活化(2h)。PCR法测TLR4mRNA及NF-?B mRNA的表达,免疫印迹(Western-blot)技术测TLR4蛋白及NF-?B蛋白的表达。结果:血清作用24h后,高血压病组TLR4 mRNA的表达较健康组增高,组间差异显著。黄芪多糖可呈剂量依赖性减少LPS诱导的TLR4 mRNA高表达,抑制LPS诱导的TLR4蛋白高表达和I?Ba蛋白的降解,组间差异显著。结论:TLR-NF-?B信号途径介导的炎症反应和免疫紊乱是高血压病血管内皮损伤的机制之一,黄芪多糖可通过抑制其表达保护血管内皮细胞损伤。

【Abstract】 Objective:The research was observe the TLR-NF-?B signal pathway,to investigate the influencing mechanisms on TLR4、NF-?B expression in damage of vascular endothelial cells of hypertension disease by the effects of APS. Methods:The human umbilical vein endothelial cells(HUVEC-Cs) were intervened by the 10%serum of the patient of hypertension disease and the healthy human separately for 24h,the TLR4mRNA expression in HUVEC-Cs was detected by real time PCR.The level of TLR4 expression(24h) and NF-?B activation(2h) in lipopolysaccharide(LPS) activated HUVEC-Cs were effected by different dosage APS during the same time.TLR4 mRNA and NF-?B mRNA were detected by real time PCR,TLR4 protein and NF-?B protein were detected by western blot.The NF-?B expression reflected withⅠ?Ba.Results:After the serum acted on the HUVEC-Cs for 24h,the HUVEC-Cs TLR4mRNA expression was increased in hypertension disease group than the healthy group,there was a significant difference.APS had the function of reduce the TLR4mRNA expression,reduce the TLR4 protein expression and suppress theⅠ?B protein degeneration in LPS activated HUVEC-Cs,there was a significant difference.Conclusion: Inflammatory reaction and immunity disorder initiated by TLR-NF-?B signal pathway is one of the influencing mechanisms in the damageof vascular endothelial cells of hypertension disease, and APS can reduce the damage.
