江苏省肠道病毒71型分离株的分子遗传进化特征

【摘要】 目的了解肠道病毒71型(enterovirus 71,EV71)江苏流行株的遗传学背景。方法 RT-PCR反应测定江苏株EV71Jiangsu.P.R.C07.0810(以下简称Jiangsu01)的全基因组序列,包括5’UTR区与编码区的核苷酸序列,利用Mega3.1软件进行序列的同源性比较和种系发生分析。结果 Jiangsu01在5’UTR区与编码区共有7 320个碱基,与其他EV71代表株相比,Jiangsu01在编码区没有碱基的插入和缺失,但在5’UTR区序列长度有一定的差异。基于VP1区核苷酸序列分析表明Jiangsu01株与中国大陆代表株SHZH98同属于EV71病毒C4亚型,相应氨基酸位点的改变也与C基因型遗传进化特点相符合。核苷酸同源性分析显示,除2B区外,Jiangsu01与SHZH98株的相似度最高(91.6%),与国际标准株BrCr株和MS/7423/87(以下简称MS)的相似性较低(分别为76.5%和79%)。而在2B区,无论是核苷酸序列同源性还是氨基酸序列同源性,Jiangsu01株与MS株的相似度均为最高(分别为77%和94.8%),提示Jiangsu01株可能存在C基因型和B基因型的重组。对该病毒潜在的功能性核苷酸位点及氨基酸位点分析表明,这些位点的改变与该病毒的流行病学资料相符合。结论通过分析EV71病毒的基因特征,从分子水平上解释该毒株引起流行的可能原因和流行特征,对该病毒所致疾病的防控具有重要指导意义。

【Abstract】 Objective To understand the genetic characteristics of EV71 strains in popular in Jiangsu province.Methods RT-PCR was used for 5’ UTR and coding region of EV71Jiangsu.P.R.C 07.08 10(Jiangsu01 strain) and Mega3.1 soft was applied for homology and phylogenetic analysis. Results The length of 5’ UTR and coding region of Jiangsu01 strain was 7 320bp.Insertion or deletion was not detected in the coding region,however,there were several insertions and deletions in 5’ U TR regions.Based on the nucleotide sequence of VP1 region,phylogenetic analysis showed that Jiangsu01 strain was high similar with SHZH98 strain,the representative strain of China,and both of them belonged to C4 genotype.The VP1 amino acid mutation analysis showed the similar result.Through the homology analysis among Jiangsu01 strain,SHZH98 strain,BrCr strain and MS/7423/87 strain(MS),the highest nucleotide similarities were found between Jiangsu01 strain and SHZH98 strain in all regions(91.6%), except for 2B region.However,in 2B region,the highest nucleotide(77%) and amino acid(94.8%) similarities were found between Jiangsu01 strain and MS strain.These rusults demonstrated the possibility of intratypic recombination had been taken place between C genotype and B genotype in Jiangsu.Analyzing the potential functional nucleotide and amino acid sites,we found that the mutation of these sites were consist with the epidemiology characters of Jiangsu01 strain.Conclusions Though analysis of the genetic characteristics and of an enterovirus 71 strain isolated in Jiangsu Province,we explained the possible reasons of the prevalence and epidemiology characters of this strain on the molecular level,which will contribute to prevention and control of EV71 diseases.
