【摘要】 目的探讨解毒祛瘀滋阴中药对MRL/1pr小鼠外周血淋巴细胞凋亡及其caspase2、caspase3蛋白的影响。方法80只MRL/1pr小鼠随机分为:模型组:20只,灌胃生理盐水;中药组:20只,灌胃解毒祛瘀滋肾中药煎剂;西药组:20只,灌胃强的松混悬液;中西药组:20只,中西药分别灌胃;正常组:昆明小鼠选用20只,生理盐水灌胃,均每日1次,每次0.5ml。连续饲养用药12周后取血,以梯度离心法分离提纯外周血淋巴细胞(PBLC),培养48小时,流式细胞仪检测凋亡率;另Western blot法检测PBLC Caspase2及Caspase3的蛋白表达。结果正常组、中药组、西药组和中西药组PBLC 0和48小时凋亡率高于模型组,差异有显著性(p<0.05,p<0.01)。0小时西药组、正常组与中西药组的凋亡率无明显差异(p>0.05),但中药组和模型组与中西药组比较,差异有显著性(p<0.05);48小时西药组与中西药组的凋亡率有明显差异(p<0.05),中药组和模型组与中西药组比较,差异有显著性(p<0.01);Caspase-2蛋白水平由高到低依次为:中西药组>西药组>中药组>正常组>模型组;Caspase-3蛋白水平被激活的由高到低依次为:西药组>中西药组>中药组>正常组>模型组。结论解毒祛瘀滋阴药对MRL/1pr小鼠的PBLC凋亡率及Caspase2、3蛋白表达均有影响,能协同糖皮质激素对PLBC凋亡障碍起调整作用。更多还原

【Abstract】 Objective To explore the adjustable effects of Jiedu Quyu Ziyin Recipe(JQZR) on the apoptosis and expressions of caspase2 and caspase3 of peripheral-blood lymphocyte in MRL/lpr mice.Methods 80 MRL/lpr mice were randomly divided into model group,TCM group, Western medicine group and TCM and Western medicine group,20 mice every group,meanwhile,20 Kunming mice were divided into normal group,then intragastrically administrated normal sodium,JQZR apozem,prednisone suspension and JQZR apozem and prednisone suspension,0.5ml every time,once daily for 12 weeks respectively.At the end of the 12th week, peripheral-blood lymphocytes of every mice purified by gradient centrifugation were cultivated for 48 hours then the apoptosis was detected by flow cytometry.Furthermore, the expressions of caspase2 and caspase3 of peripheral-blood lymphocyte were detected by Western blot.Results At Oh or 48h,the apoptosis ratios of PBLC in normal group,TCM group, Western medicine group and TCMand Western medicine group are higher than model group and the differences are significant(p<0.05,p<0.01),while,at Oh,which are not significant within Western medicine group,normal group and TCM and Western medicine group(p>0.05), even if which are significant between TCM group and TCMand Western medicine group or between model group and TCM and Western medicine group(p<0.05).At 48h,the differences of apoptosis ratios of PBLC are significant between Western medicine group and TCM and Western medicine group(p<0.05),and between TCM group and TCM and Western medicine group or between model group and TCM and Western medicine group which are significant,too(p<0.01).In descending order of Caspase-2 level,the five groups’ rank is TCMand Western medicine group,Western medicine group,TCMgroup,normal group,model group.In descending order of Caspase-3 level, the five groups’ rank is Western medicine group,TCM and Western medicine group,TCM group, normal group,model group.Conclusion JQZR has adjustable effects on the apoptosis and expressions of Caspase2 and Caspase3 of Peripheral-blood Lymphocyte in MRL/1pr Mice.更多还原
