若干气道炎症性疾病动物模型气道阻力、细胞因子与HPA轴等的观察(英文)

【摘要】 目的:建立支气管哮喘(bronchial asthma,BA)、慢性阻塞性肺疾病(chronic obstructive pulmonarydisease,COPD)、慢性阻塞性肺疾病叠加哮喘(COPD+BA)等气道炎症性疾病的动物模型并对其进行比较与分析,特别是叠加模型的部分病理生理改变和下丘脑-垂体-肾上腺(hypothalamic-pituitary-adrenal,HPA)轴的变化。方法:用挪威大鼠(BN大鼠)建立哮喘、COPD、COPD+BA等气道炎症性疾病动物模型,从大鼠行为学、肺组织病理学、气道反应性、炎性细胞因子、HPA轴的变化等方面对模型进行评价与分析。结果:与对照组相比,BA模型组、COPD模型组、COPD+BA模型组BN大鼠的行为学、肺组织病理学、气道反应性、炎性细胞因子及HPA轴等均有一定的变化。其中BA组出现典型的BA性病理生理改变;COPD组出现相应的特征性改变;而COPD+BA组则兼有两者的特征性改变,并出现某些特征性病理生理改变的弱化或强化的现象。结论:利用BN大鼠,可建立较稳定的BA、COPD、COPD+BA动物模型,通过对三种动物模型的比较、分析,发现COPD+BA兼有COPD和BA各自的特征性病理生理改变,由于两种发病机制同时启动会产生交互影响,进而弱化或强化某些特征性的病理生理改变;BA、COPD和COPD+BA均存在多水平的HPA轴功能紊乱或者低弱,其中COPD+BA组最为明显,特别在下丘脑水平。
【Abstract】 Aim:This study evaluated some animal models of inflammatory airway diseases such as bronchial asthma(BA),chronic obstructive pulmonary disease(COPD) and COPD+BA,especially in the pathophysiology of the COPD+BA model group and the changes of hypothalamic-pituitary-adrenal(HPA) axis of these three animal models. Methods:BN rats were used to model the inflammatory airway diseases of BA,COPD and COPD+BA,and these three models were compared and evaluated from rat’s ethology,lung histopathology,airway hyperresponsiveness,inflammatory cytokines and the HPA axis. Results:Comparing to the control group,the rat’s ethology,lung histopathology, airway hyperresponsiveness,inflammatory cytokines and the HPA axis of these three model groups were influenced.The rats of BA model groups demonstrated the features of relevant diseases,and the same to the rats of COPD model group.However the rats of COPD+BA model group showed the features of BA and COPD,and some of the key features of pathophysiology of this model group demonstrated the tendency of Strengthened or weakened. Conclusions:BN rat can be used to establish more stable animal models of BA,COPD and COPD+BA.Comparing and evaluating these three models may contribute to find that the rats of COPD+BA model group have demonstrated the pathophysiological features of COPD and BA,which may be associated with the interaction produced by the start-up of two different mechanisms.The HPA axis disturbances or attenuations in these three model groups especially in COPD+BA model group are obvious in rat hypothalamus.
