人RNF181基因表达的降低可以抑制人肺癌细胞系A549的增殖

【摘要】 目的 :探讨E3-泛素蛋白连接酶RNF181在肺癌中的作用。方法 :利用RT-PCR在12个非小细胞肺癌(NSCLC)细胞系中对hRNF181基因的转录水平进行检测;采用RNAi技术,建立RNF181稳定knock-down的A549细胞系,对细胞增殖及细胞周期进行检测,明确目的基因在A549细胞中的功能。结果 :RNF181在不同NSCLC细胞系中的表达存在差异;降低RNF181的表达可以抑制A549细胞的增殖。结论 :降低RNF181的表达可以抑制A549细胞的增殖。
【Abstract】 Objective:To validate the function of E3 ubiquitin protein ligase RNF181 in lung cancer.Methods:RTPCR was used to detect the transcription level of hRNF181 in 12 non -small cell lung cancer cell lines.After establishment of stably knock-down RNF181 in A549 by RNAi,cell proliferation and cell cycle distribution were examined to identify the exact function of this protein in A549 cells.Results:Expression levels of RNF181 in 12 NSCLC cell lines were different.And reduced expression of RNF181 in A549 could induce inhibitory cell proliferation.Conclusion:Reduced expression of RNF181 inhibited cell proliferation in A549 cells.
