GHS-R1a在大鼠下丘脑-垂体-卵巢轴上的定位与mRNA表达

【摘要】 目的:分析ghrelin受体(GHS-R1a)在大鼠下丘脑-垂体-卵巢轴上的定位与表达。方法:采用免疫组织化学法检测GHS-R1a的定位,实时荧光定量PCR检测GHS-R1a mRNA的表达。结果:GHS-R1a在下丘脑中主要分布在弓状核,腹内侧核,正中隆起的颗粒细胞、椎体细胞以及穿插于颗粒细胞和椎体细胞之间的神经胶质细胞内;在脑垂体中主要分布在腺垂体的嗜酸性细胞和嗜碱性细胞内,而神经垂体则无;以及卵巢的卵母细胞、黄体细胞初级卵泡的粒层细胞、有腔卵泡的内膜层和粒层细胞有少量GHS-R1a分布,在原始卵泡的粒层细胞、有腔卵泡阶段外膜层细胞没有GHS-R1a分布。GHS-R1amRNA在不同发情周期大鼠的下丘脑、垂体均有表达,下丘脑中GHS-R1a mRNA在发情后期达到最高值,发情期最低;脑垂体中,GHS-R1a mRNA发情前期最高,间情期达到最低值;大鼠卵巢中没有GHS-R1amRNA分布。结论:GHS-R1a在下丘脑-垂体-卵巢轴中均有分布。GHS-R1a mRNA在大鼠的下丘脑、垂体中的表达受发情周期的控制。

【Abstract】 Objective:To analyze the distribution and gene expression of ghrelin receptor(GHS-R1a) in hypothalamus-pituitary-ovarian axis of rat.Methods:the distribution of GHS-R1a were detected by Immunohistochemical,real-time quantitative PCR was used to detect gene expression of GHS-R1a. Results:In the hypothalamus,GHS-R1a mainly distributed in the granulosa cells,vertebral body cells and the glial cells in arcuate nucleus,ventral medial nucleus and median eminence.In the pituitary, GHS-R1a mainly located in the eosinophilic cells and basophilic cells of anterior pituitary,and no GHS-R1a immunopositive cells were observed in neurohypophysis.In the ovary,GHS-R1a was mainly distributed in the luteal cells,hilus interstitial cells,oocyte,granulosa cell of primary follicles,the theca interna and granulosa cell of antral follicle showed weak GHS-R1a immunostaining.However,we failed to find GHS-R1a in the granulosa cells of the primordial follicles and the theca externa cell of antral follicle.In addition,GHS-R1a mRNA were expressed in hypothalamus and pituitary of rat with different estrous cycle.In the hypothalamus,GHS-Rla mRNA was highest in the late estrus,and the lowest in the estrus period.In the pituitary,GHS-R1a mRNA was highest in the proestrus,but the lowest in the diestrus. GHS-R1a mRNA was no expressed in ovary of rat.Conclusion:GHS-R1a was presented in the hypothalamus-pituitary-ovarian axis of rat.The expression of GHS-R1a mRNA in the hypothalamus and pituitary was under the control of estrous cycle.
