梓醇对过氧化氢诱导的星形胶质细胞氧化损伤的保护作用

【摘要】 目的:研究梓醇对过氧化氢(H2O2)诱导的大鼠脑星形胶质细胞氧化性损伤的保护作用。方法:采用1、10和100μmol·L-1的梓醇预处理星形胶质细胞24 h后,再用300μmol·L-1 H2O2损伤细胞。倒置显微镜观察各组细胞形态变化,MTT法检测细胞存活率的改变,比色法检测细胞培养液中乳酸脱氢酶(LDH)的含量变化以及细胞内的超氧化物歧化酶(T-SOD)、谷胱甘肽过氧化物酶(GSH-PX)活性变化和丙二醛(MDA)的含量变化。结果:梓醇可明显改善细胞的形态变化,提高了星形胶质细胞的存活率,维持了细胞膜的完整性,并对H2O2诱导的星形胶质细胞内T-SOD、GSH-PX活性降低和MDA含量升高有显著的抑制作用。结论:梓醇对H2O2诱导的星形胶质细胞氧化性损伤具有保护作用。
【Abstract】 Objective:To study the protective effects of catalpol against H2O2-induced oxidative damage in astrocytes.Method:Astrocytes were pretreated with 1,10,100μmol·L-1 catalpol and then exposed to H2O2. Effects of catalpol on cell morphous were observed by inverted microscope.Cell viability were deteced by MTT assay;LDH content in the cell culture fluid and intracellular T-SOD,GSH-Px activity,MDAcontent were assessed by colorimetric method.Results:The results showed catalpol evidently ameliorated cell morphous,improved cell viability,and maintained integrality of cell membrane.Furthermore,catalpol can significantly inhibit the decrease of intracellullar T-SOD,GSH-PX activity,and the increase of MDA content. Conclusion:Catalpol have protective effects against H2O2-induced oxidative damage in astrocytes.
