板蓝根生物碱酸性部位对新城疫病毒吸附、释放的影响

【摘要】 为研究板蓝根生物碱酸性部位(RIAa)对抗新城疫病毒(NDV)的机理,本试验通过细胞病变抑制法和MTT比色法检测了RIAa对NDV感染CEF吸附和释放的影响.结果显示药毒混合方式中在60min,RIAa为31.3μg/mL时的A570值显著高于病毒对照(P<0.05),在120min时3个浓度均显著高于病毒对照(P<0.05);先加药物方式中在120min,浓度为62.5和31.3μg/mL时均显著高于病毒对照(P<0.05);先加毒吸附方式中,各吸附时间的A570值与病毒对照差异均不显著(P>0.05).RIAa+NDV的细胞上清再次作用细胞后其A570值显著高于没有加RIAa的NDV对照(P<0.05).空斑试验进一步证实了RIAa的抗NDV作用.结果表明,吸附时间、加药方式对病毒感染细胞的能力有显著影响,RIAa在一定浓度下对病毒的吸附有阻碍作用,能够保护细胞减少病毒感染,但是不能解除已经吸附的病毒;RIAa在一定浓度下能够显著抑制病毒的释放.
【Abstract】 Effect of acid of Radix Isatidis Alkaloid(RIAa) on adsorption and release of Newcastle Disease Virus(NDV) was detected by cytopathic effect inhibition and MTT colorimetric assays.The results showed that under the treatment of adding drug and virus simultaneously,the A570value at the concentration of 31.3μg/mL when absorbed 60min was significantly higher than virus control(P<0.05).However,the values of all the three concentrations when absorbed 120min were significantly higher than virus control(P<0.05).With the treatment of adding drug firstly,when absorbed 120min,the A570 values of RAIa at the concentrations of 62.5 and 31.3μg/mL were significantly higher than virus control(P<0.05).However,the A570 values of different absorbed time when the virus was added firstly showed no significant difference with virus control(P>0.05).In the experiment of adding cell supernatant into CEF again,the A570 values of RIAa+NDV were significantly higher than virus control(P<0.05). Its aitivirus ability was confirmed by the plaque formation assay.The results indicated that absorption time and the methods of adding drugs showed significant effect on the ability of virus infecting cells.RIAa at a certain concentration could inhibit virus absorbing into cells,and protect cells from infecting virus.However,it could not remove virus which had been absorbed on cells. On the other hand,RIAa could remarkablely inhibit virus’s releasing.
