HPLC-DAD-MS/MS分析芍药甘草汤大鼠血中移行组分的研究

【摘要】 目的采用HPLC-DAD-MS/MS分析芍药甘草汤大鼠给药后血浆样品,确认芍药甘草汤入血移行成分。方法以kormasil C18(4.6×250mm,带保护柱)为色谱柱;以乙腈和0.5%乙酸/水为流动相进行梯度洗脱,流速为0.8mL·min-1,柱温:25℃,以甲醇为去蛋白溶剂,样品经电喷雾离子源正离子化后,通过LCQdeca型质谱仪,在m/z100~1000范围内进行扫描,并对特征离子进行二次裂解,获得二级质谱数据。结果以芍药甘草复方组方前后的体外成分研究为基础,确认了芍药甘草汤体外的主要成分为芍药苷、白芍苷、甘草苷、甘草酸等,而入血移行成分中未发现此类糖苷类物质,却发现了来源于甘草成分的葡萄糖醛酸结合物——甘草素葡萄糖醛酸和异甘草酸葡萄糖醛酸。结论采用液相色谱-质谱联用技术分析芍药甘草汤大鼠血中移行组分,并与芍药甘草复方组方前后的成分进行对比,可对复方入血的成分进行推测和确认,具有简便、快速的优点,且对于中药复方体内代谢成分的研究具有较好的应用价值。

【Abstract】 OBJECTIVE To study the adscription of plasma effective constituents of rat after oral administration of Shaoyao-gancao decoction ( SGD ) by HPLC-DAD-MS/MS, and confirm the effective constitutes in plasma. METHODS kormasilC18 (4.6×250 mm, with protection column) was chromatographic column; acetonitrile and 0.5 % acetic acid/water was mobile phase for gradient elution, flow rate was 0.8 mL·min-1, column temperature: 25 ℃,methanol was used to remove the protein, A LCQdeca mass spectrometer system equipped with an electrospray ionization iontrap source was used as the detector and operated in the positive ion mode. Ions was scanned from the m/z 100 to m/z 1000, and characteristic ion were secondary schizolysised to obtain data of second order MS. RESULTS The main constitutes of SGD were peoniflorin, Albiflorin R1, Liquiritin and glycyrrhizic acid and so on, but no glucosides was found in the plasma samples of rats administered with SGD, glucuronide conjunctions of liquiritigenin and isoliquiritigenin were detected in plasma sample. CONCLUSION liquid chromatography-mass spectrometry technology provides a convenient and high speed method to analyze the effective constitutes of plasma of rats administered with SGD, therefore, it has a good value in studies on the metabolism of traditional Chinese medicine ( TCM ).
