肿瘤坏死因子α基因-238G>A及-857C>T多态性与河南汉族群体原发性高血压

【摘要】 目的探讨肿瘤坏死因子α(TNF-α)基因-238G>A及-857C>T多态性与河南汉族原发性高血压(EH)的相关性。方法ELISA法检测130例高血压患者和197健康对照者血浆TNF-α水平,采用限制性扩增片段长度多态性(PCR-RFLP)的方法检测TNF-α基因-238G>A及-857C>T多态性。结果与正常对照组相比,高血压患者血浆TNF-α水平显著增高(P<0.01);EH组中TNF-α-238G>AGA+AA基因型者的DBP、TC、TG、及LDL含量明显高于GG纯合子者,差异有统计学意义(P<0.05),但是EH组TNF-α基因-238G>A及-857C>T各基因型及等位基因频率与正常对照组比较差异无统计学意义(P>0.05)。结论TNF-α-238G>AA等位基因可能会增加高血压的发病风险,但TNF-α基因-238G>A及-857C>T基因多态性与河南汉族原发性高血压无关联。
【Abstract】 Objective To investigate the correlation between tumor necrosis factor α gene polymorphisms and Henan Han Essential hypertension (EH). Methods The levels of TNF-α in plasma was detected by ELISA. The gene polymorphisms of TNF-α-238G>A and -857C>T in 130 EH patients (EH group) and 197 healthy population (control group) were analyzed by polymerase chain reaction and restriction endonuclease technology (PCR-RFLP). Results In EH group, the DBP, TC, TG, and LDL levels of the TNF-α-238G> A GA + AA genotype people were significantly higher than that of GG genotype (P <0.05). However, there was no significant difference in genotype and allelic frequency of the -238G>A and -857C>T polymorphism between healthy subjects and patients with EH (P > 0.05) . Conclusion TNF-α-238G> A A allele might increase the risk of etiology in EH, but the gene polymorphism of TNF-α-238G>A and TNF-α-857C>T did not to be associated with EH in Henan Han population.
