蝙蝠葛酚性碱对大鼠局灶性脑缺血再灌注损伤的保护作用

【摘要】 目的研究蝙蝠葛酚性碱对大鼠局灶性脑缺血再灌注损伤的保护作用。方法线栓法制备大鼠大脑中动脉阻断局灶性脑缺血模型,缺血2h后再灌注24h,观察蝙蝠葛酚性碱(5、10、20mg·kg-1,再灌开始时ip给药)对神经功能评分、脑梗死范围、脑水肿和血脑屏障通透性的影响,测定脑组织中超氧化物歧化酶(SOD)活性、丙二醛(MDA)水平和谷胱甘肽过氧化物酶(GSH-Px)活性。结果蝙蝠葛酚性碱能明显减轻大鼠的神经功能缺陷、缩小脑梗死体积、减轻脑水肿和血脑屏障通透性,显著增加SOD和GSH-Px活性,降低MDA水平。结论蝙蝠葛酚性碱对脑缺血再灌注损伤有保护作用,其机制可能与降低血脑屏障通透性、抗脂质过氧化有关。

【Abstract】 Objective To study the protective effects of phenolic alkaloids from Menispermum dauricum(PAMD) on focal cerebral ischemia-reperfusion injury in rats. Methods Reversible middle cerebral artery occlusion(MCAO) model was produced by intraluminal suture technique, and reperfusion was begun after 2 h of occlusion. PAMD was ip while reperfusion begun. After 24 h reperfusion the neurological deficit score, cerebral infarct volume, brain edema and the permeability of blood brain barrier were estimated. The activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), and the level of malondialdehyde (MDA) in the brain tissue were measured. Results PAMD could significantly reduced neurological deficit, cerebral infarct volume, brain edema and the permeability of blood brain barrier. It could also increase the activities of SOD and GSH-Px, and decrease the level of MDA in the brain tissue significantly. Conclusion PAMD has protective effects on cerebral ischemia-reperfusion injury in rats. The mechanisms may be related to reducing the permeability of blood brain barrier and lipid peroxidation.
