秋水仙素诱导‘凤丹’牡丹多倍体的初步研究

【摘要】 多倍体育种是观赏植物延长花期、提高抗性的有效途径之一,在牡丹育种中尚属空白。本文采用不同浓度的秋水仙素作为诱变剂、1%二甲基亚砜作为渗透剂,对‘凤丹’牡丹混合芽与离体胚进行不同时间的诱变处理,主要结果包括:混合芽正常生长、萎蔫与死亡的比例,受秋水仙素浓度(0.1、0.2与0.5%)和处理时间(1、2与3d)的双重影响,诱变率在0.2%秋水仙素处理3d时效果最佳(35%);诱变叶片不仅细胞染色体加倍成为4倍体(2n=4x=20),而且表现出巨大性,其叶宽约为正常叶片的二倍。在培养基中直接加入0.05、0.1与0.2%的秋水仙素处理时,所有离体胚先后全部死亡;而用不同浓度(0.1、0.2与0.5%)秋水仙素、采用包裹法处理离体胚苗时,诱变率随处理时间(1、2与3d)增而提高,在浓度为0.5%、处理时间为3d时最高(25%)。
【Abstract】 Polyploid breeding,an effective approach through which the florescence of ornamental plants could be prolonged and the resistance could be improved,hasn’t been used in the breeding of tree peonies.This paper reveals our research on mutagenesis in buds and in vitro embryos of Paeonia ostii‘Feng Dan’,using colchicine as the mutagen mixed with 1%dimethylsulfoxide as the penetrant.Various concentrations of colchicine and time lengths for the treatments were tested and main results are as followed.The proportion of normally growing,wilting and died buds was influenced dually by both the concentration (0.1,0.2,0.5%) and the treating time(1,2,3 d) of colchicines and the optimal mutation(35%) was induced by the treatment of 0.2%colchicine for 3 days.The leaves in mutants weren’t twice morphologically in width as the normal leaves, but the chromosomes were doubled from diploid to tetraploid(2n=4x=20).0.05,0.1 and 0.2%colchicines added directly into the medium resulted in the death of all in vitro cultured embryos,but,in the wrapping treatments of cotton dip in0.1,0.2 and 0.5%colchicines,the mutsgenesis rate increased with the extending wrapping days(1,2,3 d) and the highest mutagenesis (25%) was obtained in the treatment of 0.5%colchicine for 3 days.
