无核葡萄‘喜乐’胚挽救培养与育种研究

【摘要】 胚挽救培养自创立至今得到了蓬勃的发展和广泛的应用,无核葡萄的胚培养方面已有了很多成功的报道,但无核葡萄胚珠培养的难易与胚珠残痕的大小密切相关,小败育型胚珠的胚挽救成功率较低。本文以小败育型的‘喜乐’葡萄杂交胚珠为材料,进行胚挽救技术研究及杂交后代无核性状的观察。结果表明,‘喜乐’胚珠接种适期大致应在盛花后30-40天左右;用二次果的胚培养成功率明显比一次果高;‘喜乐’采用1/2MS无激素浅层液体及剖胚培养等方法,既提高了胚培效率且试验结果比较稳定;以‘喜乐’为亲本的杂交后代,其无核株率平均达60.4%,以上充分验证了胚挽救技术在无核葡萄育种实践中的价值。
【Abstract】 The technician of embryo rescue was obtained flourishing development and used widely from the technique was establishment.There were many successful reports about embryo culture in seedless grape culture.The difficult degree was related to size of ovule,small abortion ovule was difficult used for embryo culture.This paper studies the technique of embryo rescue and observed the character of hybridize progenies using Himrord seedless.The results showed that the best inoculation period of was thirty to forty days after full-bloom stage and it was easier successful used second fruit than first fruit.It can be not only improving efficiency of embryo culture but also received stable results using 1/2MS liquid medium without hormone. The seedless rate of hybrid progency of"Himrord seedless"obtained mean 60.4%which show that the applying value of embryo technique in seedless grape breeding.
