苹果α-法尼烯合酶基因序列变异的进一步研究(英文)

【摘要】 α-法尼烯合酶是一种催化α-法尼烯合成的酶。在以前的研究中,我们发现了4个氨基酸序列变异,其中一个在保守的RxR序列模式区。本研究找出了其它一些序列保守区,发现另一个氨基酸序列变异(103 E→K)也在一个序列保守区。另外,我们还研究了α-法尼烯合酶基因的内含子3,结果表明内含子3的大小是1089bp(GenBank登录号FJ2263961)。当虎皮病敏感的‘国光’苹果α-法尼烯合酶基因的内含子3序列与较抗病的‘金帅’苹果有关序列比较时,在两序列间发现了12个单核苷酸变异。而在我们以前的研究中,当‘国光’苹果α-法尼烯合酶基因序列与GenBank中的一个cDNA序列(登录号AY523409)比较时,在1700 bp长的外显子序列上只发现了6个单核苷酸变异,这说明内含子3是一个高变异区。

【Abstract】 Alpha-farnesene synthase(AFS) is an enzyme which catalyzes the formation of E,E-alpha farnesene,an acyclic sesquiterpene which was considered to be related to the development of apple superficial scald.In our previous study,4 amino acid mutations were identified and one of them was found in the conserved RxR motif.Here,some other conserved regions of AFS gene were found,another amino acid mutation(103 E→K) was also found in a conserved region.The intron 3 ofAFS gene was also studied,the result showed that the size of intron 3 was 1089bp(GenBank accession No.FJ2263961).When intron 3 sequence of AFS gene of scald-susceptible cultivar‘Ralls’ was compared to that of scald-resistant cultivar‘Golden Delicious’,12 single nucleotide polymorphisms(SNPs) were identified between two sequences. While in our previous study,only 6 SNPs were found in over 1700 bp of exon sequences when the exon sequences of‘Ralls’ apple AFS gene were compared to the cDNA in the GenBank(accession No.AY523409), so the intron 3 was a highly variable region.
