NF-κB和C/EBPβ在香烟烟雾凝集物诱导BEAS-2B细胞IL-8表达中的作用

【摘要】 目的:探讨NF-κB和C/EBPβ在香烟烟雾凝集物(cigarette smokeextracts,CSE)诱导人气管上皮细胞BEAS-2B表达IL-8表达中的作用。方法:用CSE作用于不同基因型的人气管上皮细胞,并通过与IL-8促进子相连的荧光素酶(luciferase,fLCF)基因表达量的变化,观察NF-κB和C/EBPβ转录因子在CSE引起的支气管上皮细胞IL-8表达变化中的作用。结果:CSE作用三种不同基因类型BEAS-2B细胞后,IL-8WTBEAS-2B细胞fLCFmRNA表达量是IL-8mNF-κB-BEAS-2B细胞fLCFmRNA表达量的1.32倍,是IL-8mC/EBPβ-BEAS-2B细胞的fLCFmRNA表达量的1.54倍;相同浓度和作用时间的CSE刺激,IL-8mNF-κB-BEAS-2B细胞和IL-8mC/EBPβ-BEAS-2B细胞的荧光素酶活性都较IL-8WTBEAS-2B细胞低,且差异有显著统计学意义(P<0.001)。结论:NF-κB和C/EBPβ转录因子共同参与CSE诱导的IL-8基因转录活化的细胞内信号传导过程。提示NF-κB和C/EBPβ转录因子通过参与炎症因子IL-8表达,促进吸烟导致的呼吸系统炎症反应。
【Abstract】 Objective: To study effect of NF-κB and C / EBPβ on the IL-8 Expression in BEAS-2Bcells,bronchial epithelial cells, Methods: After applying CSE on different genotypes of bronchialepithelial cells,the expression of luciferase (fLCF) linked to IL-8 promoter were investigated; thus theeffect of NF-κB and C / EBPβ transcription factor on the IL-8 expression of bronchial epithelial cellsinduced by CSE were observed. Results: After applying CSE on three different genetic types ofBEAS-2B cells, fLCF mRNA expression of IL-8 WT BEAS-2B cells was 1.32 times that ofIL-8mNF-κB-BEAS-2B cells, and was 1.54 times that of IL-8mC / EBPβ-BEAS-2B cells. Under thesame concentration and time of CSE stimulation, luciferase activity of IL-8mNF-κB-BEAS-2B cells andIL-8mC/EBPβ- BEAS-2B cells were lower than that of IL-8 WT BEAS-2B cells is low, and the difference was statistically significant (P <0.001). Conclusion: NF-κB and C / EBPβ transcriptionfactors may participate in IL-8 gene transcription activation of intracellular signal transduction processinduced by CSE. It suggests NF-κB and C / EBPβ transcription factor may promote smoking-inducedinflammation of respiratory system by involving in IL-8 expression.
